药食两用药材护胃有效成分筛选及在草本白酒中的功效验证OA
Screening of gastric-protective active components in medicinal herbs with dual food and medicinal uses and validation of their efficacy in herbal spirits
筛选桔梗提取物、芦荟冻干粉、茯苓多糖三种药食两用药材相关成分中对酒精性胃损伤具有保护作用的有效成分,并验证其在草本白酒中的护胃功效.首先采用体外酒精性胃黏膜细胞损伤模型,对三种成分的护胃活性进行筛选;随后以SPF级SD大鼠为研究对象,建立慢性酒精性胃黏膜损伤大鼠模型,将筛选得到的有效成分添加至 53 度草本清香酒体中,通过检测大鼠体重、胃黏膜损伤指数、胃组织生化指标(EGF、PGE2、MDA、GAS、α1 酸性糖蛋白、α2 巨球蛋白)及病理形态,验证其护胃功效.结果如下:体外细胞实验显示,桔梗皂苷D(桔梗提取物主要活性成分)、芦荟冻干粉、茯苓多糖对酒精性胃细胞损伤具有保护作用;体内动物实验表明,53 度草本清香酒体中添加桔梗提取物(200~400 mg/L)、芦荟冻干粉(100 mg/L)、茯苓多糖(100 mg/L)后,可显著降低大鼠胃黏膜损伤积分,调节胃组织生化指标趋于正常,改善胃黏膜病理损伤,且对大鼠体重无明显不良影响.因此,桔梗提取物、芦荟冻干粉、茯苓多糖是具有良好护胃潜力的药食同源成分,可有效缓解酒精诱导的慢性胃黏膜损伤,适宜在草本白酒中添加应用.
In order to screen active components with protective effects against alcoholic gastric injury from three medicinal and edible herb-related ingredients,namely Platycodon grandiflorum extract,aloe freeze-dried powder,and Poria cocos polysaccharide,and to verify their gastric protective efficacy in herbal spirits;firstly,an in vitro alcoholic gastric mucosal cell injury model was used to screen the gastric protective activities of the three components.Subsequently,SPF-grade SD rats were used as research objects to establish a chronic alcoholic gastric mucosal injury model.The screened active components were added to 53%(alcohol by volume,ABV)Qingxiang NianDai base wine.The gastric protective efficacy was verified by detecting rat body weight,gastric mucosal injury index,gastric tissue biochemical indicators(EGF,PGE2,MDA,GAS,α1-acid glycoprotein,α2-macroglobulin)and pathological morphology.The in vitro cell experiments showed that platycodin D(the main active component of Platycodon grandiflorum extract),aloe freeze-dried powder,and Poria cocos polysaccharide had protective effects on alcoholic gastric cell injury.In vivo animal experiments showed that adding Platycodon grandiflorum extract(200-400 mg/L),aloe freeze-dried powder(100 mg/L),and Poria cocos polysaccharide(100 mg/L)to 53%ABV Qingxiang NianDai wine could significantly reduce the gastric mucosal injury score of rats,regulate the gastric tissue biochemical indicators to tend to be normal,improve the pathological injury of gastric mucosa,and had no significant adverse effect on the body weight of rats.In conclusion,Platycodon grandiflorum extract,aloe freeze-dried powder,and Poria cocos polysaccharide were medicinal and edible components with good gastric protective potential,which could effectively alleviate alcohol-induced chronic gastric mucosal injury.They are suitable for addition and application in herbal spirits.
