基于胃蛋白酶消化作用的高通量筛选方法建立及其对200种中药材的活性评价OA
Establishment of a high-throughput screening method based on pepsin digestion and its application in bioactivity evaluation of 200 traditional Chinese medicines
建立一种基于胃蛋白酶能促进消化的机理的高通量筛选方法,以便为快速筛选具有促进消化功能的中草药提供筛选依据.以酪氨酸为阳性对照,采用多功能酶标仪及微孔板为载体,通过反应条件优化,对200种中药提取液进行筛选评价.200种中药材中清热药、解表药中高活性的比例较高;而补益药、矿物药多表现出较低的活性.基于胃蛋白酶的促进作用建立的高通量筛选方法具有稳定、高效、低成本,可用来快速筛选中药材及天然成分,为中药材促进消化功能的研发提供参考.
A high-throughput screening method based on the promoting effect of pepsin was established to provide a scientific basis for rapid identification of digestion-enhancing Chinese herbal medicines.Using tyrosine as the positive control,a multifunctional microplate reader and 96-well microplates were employed as the experimental platform.After optimization of reaction conditions,200 extracts from Chinese medicinal herbs were systematically screened and evaluated.Among these 200 species,heat-clearing herbs and exterior-resolving herbs exhibited higher proportions of high activity,while tonifying herbs and mineral-based herbs predominantly showed low activity.The established screening method demonstrated stability,high efficiency,and cost-effectiveness,making it suitable for rapid screening of Chinese medicinal herbs and natural compounds.This approach provides a valuable reference for the development and research of herbal medicines targeting digestive function improvement.
