三种蜜环菌子实体对氯苯丙氨酸诱导失眠小鼠的影响OA
Sedative Effects of Fruiting Bodies of Three Armillaria Species on p-Chlorophenylalanine-Induced Insomnia in Mice
分别采用高卢蜜环菌(Armillaria gallica)、奥氏蜜环菌(A.ostoyae)和沙地蜜环菌(A.desertorum)子实体粉末(高、低剂量分别为2.0、1.0 g·kg-1)灌胃对氯苯丙氨酸(para-chlorophenylalanine,PCPA)诱导的失眠小鼠,测定其体质量及Morris水迷宫逃避潜伏期(Morris water maze escape latency),并检测血清中白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)和海马组织中 5-羟色胺(5-hydroxytryptamine,5-HT)、γ-氨基丁酸(γ-aminobutyric acid,GABA)、去甲肾上腺素(norepinephrine,NE)、多巴胺(dopamine,DA)、谷氨酸(glutamic acid,Glu)含量,观察苏木素-伊红染色小鼠肝脏、心脏、海马组织显微结构.结果表明:与模型组相比,各给药组小鼠体质量均显著增加且逃避潜伏期均显著缩短.供试3种蜜环菌均能降低小鼠血清中IL-6、IL-1β和TNF-α含量;提高小鼠海马组织中5-HT、GABA含量,降低小鼠海马组织中NE、DA和Glu含量;同时改善小鼠失眠导致的肝脏、心脏、海马组织损伤,研究结果可为蜜环菌属真菌进一步开发利用提供参考.
The effects of three Armillaria species—Armillaria gallica,A.ostoyae,and A.desertorum—on para-chlorophenylalanine(PCPA)-induced insomnia were studied in a mouse model.Mice were intragastrically administered fruiting body powders of the three species at high(2.0 g·kg-1)and low(1.0 g·kg-1)doses.Body mass and Morris water maze escape latency were measured.Serum levels of interleukin-6(IL-6),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α),as well as hippocampal levels of 5-hydroxytryptamine(5-HT),γ-aminobutyric acid(GABA),norepinephrine(NE),dopamine(DA),and glutamic acid(Glu),were determined.Histopathological changes in the liver,heart,and hippocampus were observed using hematoxylin-eosin(HE)staining.The results showed that,compared with the model group,all treatment groups showed significant increases in body mass and significant reductions in escape latency.All three Armillaria species decreased the levels of IL-6,IL-1β and TNF-αin serum.They elevated hippocampal levels of 5-HT and GABA while decreasing NE,DA and Glu levels.They also ameliorated insomnia-induced damage to the liver,heart and hippocampus.These findings provide a scientific reference for the further development and utilization of Armillaria species.
