集约化培养香菇菌棒的理化指标及其相关性OA
Respiration Intensity,Physicochemical Indicators,and Their Correlation in Lentinula edodes Cultivation Sticks During Intensive Cultivation
为探讨集约化培养香菇(Lentinula edodes)菌棒的理化指标及其相关性,分别测定培养过程中菌棒的呼吸强度、含水量、pH,葡萄糖、D-木糖、有机碳、全氮、纤维素、半纤维素、木质素、乙醇、乳酸和ATP含量,以及羧甲基纤维素酶、木聚糖酶、漆酶和Na+/K+-ATP酶活性,并分析这些理化指标间的相关性.结果表明:随着培养时间增加,呼吸强度先增加后下降,在培养43 d(第1次刺孔后2d)时达到最大值;含水量、pH、碳氮比和纤维素、半纤维素、木质素含量在培养88 d时均比10 d时低,D-木糖和乙醇含量在培养40 d时达到最大值,葡萄糖、乳酸含量和羧甲基纤维素酶、木聚糖酶、漆酶活性在培养88 d时达到最大值,ATP含量和Na+/K+-ATP酶活性在培养42 d时达到最大值;呼吸强度与Na+/K+-ATP酶活性、ATP含量存在显著正相关(r≥0.79,P<0.05),与pH存在显著负相关(r=-0.72,P<0.05).本研究结果可为香菇集约化培养提供参考.
To investigate the respiration intensity,physicochemical indicators,and their correlation in Lentinula edodes cultivation sticks(inoculated bags filled with culture substrate)under intensive cultivation,the following parameters of the cultivation sticks were measured throughout the cultivation process:respiration intensity,moisture content,pH,glucose content,D-xylose content,organic carbon content,total nitrogen content,cellulose content,hemicellulose content,lignin content,ethanol content,lactic acid content,ATP content,carboxymethyl cellulase activity,xylanase activity,laccase activity,and Na+/K+-ATPase activity.The correlations among these indicators were also analyzed.The results showed that respiration intensity initially increased and then decreased,reaching its maximum at 43 d of cultivation(2 d after the first aeration treatment by punching holes on the cultivation bags).By 88 d of cultivation,moisture content,pH,C/N ratio,and the contents of cellulose,hemicellulose,and lignin were all lower than those at 10 d.The contents of D-xylose and ethanol peaked at 40 d of cultivation,while the contents of glucose and lactic acid,along with the activities of carboxymethyl cellulase,xylanase,and laccase,reached their highest levels at 88 d of cultivation.ATP content and Na+/K+-ATPase activity peaked at 42 d of cultivation.Respiration intensity showed a significant positive correlation with Na+/K+-ATPase activity and ATP content(r≥0.79,P<0.05),and a significant negative correlated with pH(r=-0.72,P<0.05).These findings provide a reference for the intensive cultivation of L.edodes.
