利用NCⅡ(North Carolina Ⅱ)试验设计,选取'申香215'的8个孢子单核体菌株与8个可交配的孢子单核体菌株进行不完全双列杂交,得到64个杂交组合.测定杂交后代在PDA平板上和木屑培养基中的菌丝生长速度,分析孢子单核体菌株的一般配合力、杂交组合的特殊配合力和菌丝生长速度的遗传方差成分.结果表明:供试菌株间的菌丝生长速度差异极显著;孢子单核体菌株的菌丝生长速度与杂交后代在PDA平板上的菌丝生长速度无显著相关性;杂交后代在PDA平板上和在木屑培养基中的菌丝生长速度极显著相关;供试孢子单核体菌株间的一般配合力及杂交组合的特殊配合力差异均极显著;杂交后代在PDA平板上菌丝生长速度的遗传效应以加性效应为主,狭义遗传力为63.18%;在木屑培养基中的菌丝生长速度受遗传背景及培养环境共同影响,狭义遗传力为38.97%;孢子单核体菌株的一般配合力与其杂交组合的特殊配合力无相关性;孢子单核体菌株M4-4、M2-2、Csm82、Csm89的一般配合力较高,可作为骨干亲本用于选育菌丝生长速度快的新种质.

Using a North Carolina Ⅱ(NC Ⅱ)experimental design,eight spore-derived monokaryotic strains isolated from the Lentinula edodes cultivar'Shenxiang 215'were crossed with eight compatible spore-derived monokaryotic strains through an incomplete diallel cross,yielding 64 hybrid combinations.Mycelial growth rates of the hybrid progeny were measured on both potato dextrose agar(PDA)plates and sawdust-based medium.General combining ability(GCA)of the spore-derived monokaryotic strains,specific combining ability(SCA)of the hybrid combinations,and genetic variance components of mycelial growth rate were analyzed.The results showed extremely significant differences in mycelial growth rate between the tested strains.There was no significant correlation between the mycelial growth rates of the spore-derived monokaryotic strains and that of their hybrid progeny on PDA plates.However,mycelial growth rates of the hybrids on PDA plates and sawdust medium were extremely significantly correlated.Both the GCA between the tested spore-derived monokaryotic strains and the SCA between the hybrid combinations were extremely significantly different.The genetic effects governing mycelial growth rate on PDA plates were predominantly additive,with a narrow-sense heritability of 63.18%.In contrast,mycelial growth on sawdust medium was affected by both genetic background and culture environment,with a narrow-sense heritability of 38.97%.There was no correlation between the GCA of the spore-derived monokaryotic strains and the SCA of their corresponding hybrid combinations.Four spore-derived monokaryotic strains,M4-4,M2-2,Csm82,and Csm89,showed relatively high GCA values,suggesting their potential as elite parent strains for breeding new germplasm with improved mycelial growth.
