高效液相色谱法同时测定西河柳叶中5种黄酮类成分含量OA

Simultaneous determination of five flavonoids in Tamaricis cacumen by HPLC

为了建立一种可同时测定西河柳叶中槲皮素、木犀草素、山奈酚、柽柳黄素和山柰素含量的高效液相色谱分析方法,采用Shim-pack GIST C18(4.6 mm×150 mm,5 μm)色谱柱,以甲醇-0.01%磷酸氢二钾溶液(梯度洗脱)为流动相,流速为 0.5 mL/min,检测波长为 366 nm,柱温为 35℃,进样量为 10 μL,测定西河柳中槲皮素、木犀草素、山奈酚、柽柳黄素和山柰素含量.结果显示槲皮素、木犀草素、山奈酚、柽柳黄素和山柰素在 0.0017~0.1632 mg/mL 范围内与峰面积呈现良好的线性关系(R2≥0.9987);其精密度的RSD≤1.8%;重复性 RSD≤1.8%;槲皮素、木犀草素、山奈酚、柽柳黄素和山柰素的回收率在95%~105%且 RSD≤2.0%,样品在室温下放置 12 h 稳定性良好.本研究建立了西河柳叶中 5 种黄酮类成分的含量测定方法,该方法简便、灵敏、准确、专属性强,重复性好,符合2020版药典相关标准,满足分析要求.

In order to establish a high-performance liquid chromatography(HPLC)analytical method for the simultaneous determination of quercetin,luteolin,kaempferol,tamarixin and kaempferide contents in Tamaricis cacumen leaves,the quercetin,luteolin,kaempferol,tamarixin and kaempferide contents in Tamaricis cacumen leaves were determined on a Shim-pack GIST C18(4.6 mm×150 mm,5 μm)with methanol-0.01%K2HPO4 solution(gradient elution)as the mobile phase at a flow rate of 0.5 mL/min,a detection wavelength of 366 nm,and a column temperature of 35℃with a 10 μL injection volume.The results showed that the quercetin,luteolin,kaempferol,tamarixin and kaempferide showed good linear relationship with peak area in the range of 0.0017~0.1632 mg/mL(R2≥0.9987);the RSD values of their precision were no more than 1.8%;the RSD values of their reproducibility were no more than 1.8%;the spiked recoveries of quercetin,luteolin,kaempferol,tamarixin and kaempferide were in the range of 95%~105%with RSD no more than 2.0%.The samples had good stability when placed at room temperature for 12 h.In conclusion,this study established a method for the simultaneous determination of five flavonoids in Tamaricis cacumen leaves.The method demonstrated simplicity,high sensitivity,accuracy,strong specificity,and good repeatability,complying with the relevant standards of the 2020 Pharmacopoeia and satisfying analytical requirements.
