花椒粕中绿原酸提取物的制备及其抗氧化活性研究OA
Study on preparation and antioxidant activity of chlorogenic acid extract from Zanthoxylum bungeanum residue
立足农业废弃物花椒粕高值化利用与绿原酸开发需求,以提取花椒油后的花椒粕为试验材料,建立溶剂法提取绿原酸的最佳工艺:提取温度 60℃、乙醇体积分数 80%、料液比 1∶4,此条件下绿原酸萃取得率达 94.46%.经 D101 树脂纯化后,得到绿原酸含量为 10.82%的产品.从 ABTS 自由基清除效果、金属离子螯合能力和总抗氧化能力三个维度评价显示,花椒粕绿原酸具有优秀的体外抗氧化活性.斯皮尔曼和肯德尔相关性分析表明,花椒粕中的绿原酸、没食子酸、鞣花酸与 ABTS 自由基清除、金属离子螯合和总抗氧化能力均具有较强的正相关关系,其中绿原酸与 ABTS 自由基清除能力和总抗氧化能力的相关性尤为显著.本研究为花椒粕高值化应用提供了明确的技术参数与理论依据,也为天然抗氧化剂开发提供了优质候选资源.
Based on the high-value utilization of agricultural waste—Zanthoxylum bungeanum residue and the development needs of chlorogenic acid,zanthoxylum seed cake(the by-product after Zanthoxylum oil extraction)was used as the experimental material to establish the optimal process for extracting chlorogenic acid via the solvent method.The optimal extraction conditions were determined as follows:an extraction temperature of 60℃,an ethanol volume fraction of 80%,and a solid-to-liquid ratio of 1:4.Under these conditions,the extraction yield of chlorogenic acid reached 94.46%.After purification with D101 macroporous resin,a product with a chlorogenic acid content of 10.82%was obtained.Evaluation from three dimensions—ABTS free radical scavenging effect,metal ion chelating ability,and total antioxidant capacity—showed that chlorogenic acid from Zanthoxylum seed cake exhibited excellent in vitro antioxidant activity.Spearman and Kendall correlation analyses indicated that chlorogenic acid,gallic acid,and ellagic acid in Zanthoxylum seed cake all had strong positive correlations with ABTS free radical scavenging,metal ion chelating,and total antioxidant capacity.Among them,the correlations between chlorogenic acid and ABTS free radical scavenging capacity,as well as between chlorogenic acid and total antioxidant capacity,were particularly significant.This study provides clear technical parameters and a theoretical basis for the high-value application of Zanthoxylum seed cake,and also offers a high-quality candidate resource for the development of natural antioxidants.
