新型甜味剂紫苏葶的系统性毒理学评价及安全性研究OA

研究通过急性、亚急性及遗传毒性试验,综合评价紫苏葶的安全性.结果显示,急性经口毒性试验中紫苏葶的 LD50 为 765.242 mg/kg 为低毒.亚急性紫苏葶经口毒性试验未显示明显毒性.在体外遗传毒性试验中,高质量浓度(1000 μg/皿)紫苏葶对大肠杆菌WP2 UvrA 可能具有潜在致突变性,但对其他四种鼠伤寒沙门氏菌无致突变性.在体内遗传毒性试验中,紫苏葶以 1/2 LD50、1/4 LD50 或 1/8 LD50 的剂量口服,均未引起精原细胞染色体畸变.由此可知紫苏葶整体安全性较好,但在高质量浓度(1000 μg/皿)的体外细菌回复突变试验中对大肠杆菌WP2 UvrA表现出潜在致突变性,提示需关注其高剂量下的生物学效应.

In order to comprehensively assess the safety of perillartine,this study performed acute,subacute and genotoxicity tests.The results showed that:the acute oral LD50 of perillartine was 765.242 mg/kg,which was classified as low toxicity.No significant toxicity was observed in the 28-day subacute study.In the bacterial reverse mutation assay,perillartine showed potential mutagenicity toward Escherichia coli WP2 UvrA at a high concentration(1000 μg/plate)but was not mutagenic to the four Salmonella typhimurium strains tested.In the mouse spermatogonial assay,no increase in chromosomal aberrations was found at doses of 1/2,1/4,or 1/8 of the LD50.In conclusion,the overall safety profile of perillartine was considered to be favorable under the experimental conditions of this study.However,its potential mutagenicity towards Escherichia coli WP2 UvrA at high concentrations in vitro warrants attention regarding its biological effects at high doses.
