'琐琐'葡萄对聚乙二醇模拟干旱胁迫的形态与生理响应OA
Morphological and Physiological Responses of'Suosuo'grape to PEG Simulated Drought Stress
为明确'琐琐'葡萄作为优良抗旱性砧木的理论依据,本文对聚乙二醇(Polyethylene glycol,PEG)模拟干旱胁迫下'琐琐'葡萄试管苗的形态与生理响应进行了探究.以'琐琐'葡萄试管苗为试材,通过在培养基中添加不同质量浓度的PEG-6000模拟干旱胁迫,测定旱害指数、细胞膜透性、相对含水量、渗透调节物质、丙二醛及抗氧化酶活性等指标,并进行综合评价.结果表明,随着PEG质量浓度增加与胁迫时间延长,相对电导率、丙二醛、脯氨酸、可溶性糖持续上升,相对含水量显著下降,可溶性蛋白呈先降后升再降趋势,3三种抗氧化酶(SOD、POD、CAT)活性均呈先升后降的趋势,且各胁迫处理间存在差异.基于灰色关联度分析、主成分分析和相关性分析,筛选出相对电导率、丙二醛、脯氨酸、可溶性糖和相对含水量5项指标,构成'琐琐'葡萄抗旱性综合评价体系,其中相对电导率与旱害指数的关联度最高.综上,'琐琐'葡萄在干旱胁迫下具有显著的胞膜稳定性和渗透调节能力,具备作为优良抗旱砧木的开发潜力.
In order to clarify the theoretical basis of'Suosuo'grape as an excellent drought-resistant rootstock,the morphological and physiological responses of'Suosuo'grape test-tube seedlings under polyethylene glycol(PEG)simulated drought stress were investigated.The test-tube seedlings of'Suosuo'grape were used as test materials,and different mass concentrations of PEG-6000 were added to the medium to simulate drought stress.The drought injury index,cell membrane permeability,relative water content,osmotic adjustment substances,malondialdehyde and antioxidant enzyme activity were measured and comprehensively evaluated.The results showed that with the increase of PEG concentration and the prolongation of stress time,the relative conductivity,malondialdehyde,proline and soluble sugar continued to rise,the relative water content decreased significantly,the soluble protein decreased first,then increased and then decreased,and the activities of three antioxidant enzymes(SOD,POD and CAT)increased first and then decreased,and there were differences among different stress treatments.Based on grey correlation analysis,principal component analysis and correlation analysis,five indexes of relative conductivity,malondialdehyde,proline,soluble sugar and relative water content were selected to constitute the comprehensive evaluation system of drought resistance of'Suosuo'grape,among which the correlation between relative conductivity and drought injury index was the highest.In summary,'Suosuo'grape has significant cell membrane stability and osmotic adjustment ability under drought stress,and has the potential to be developed as an excellent drought-resistant rootstock.
