两种酿酒葡萄对低温胁迫的生理响应及转录组分析OA
Physiological Response and Transcriptome Analysis of Two Kinds of Wine Grape Varieties to Low Temperature Stress
为筛选抗寒性强的酿酒葡萄种质并解析其抗寒机制,本研究以'黑比诺'(Pinot Noir)和'芳堤娜'(Frontenac)一年生枝条为试材,设4℃对照与﹣20℃低温胁迫24 h处理,测定相对电导率(REC)、可溶性糖(SS)、过氧化物酶(POD)活性,并利用转录组测序解析其抗寒机制.结果显示,低温胁迫下,两品种REC、SS含量及POD活性均升高;转录组分析中,'芳堤娜'上下调差异基因(601/610个)分布均衡,'黑比诺'下调基因占比更高(上调/下调:348/930个);GO富集显示'芳堤娜'差异基因集中于苯丙烷代谢,'黑比诺'集中于乙烯响应;KEGG富集均以代谢途径为主,且存在品种特异性;此外,MYB家族转录因子在两品种差异基因中富集程度最高,且8个基因的qRT-PCR验证结果与转录组数据高度一致,证实测序结果可靠.本研究从生理与分子层面揭示了不同抗寒性酿酒葡萄品种应对低温胁迫的差异机制,为酿酒葡萄在内蒙古地区的抗寒栽培管理提供理论依据.
To screen wine grape germplasm with strong cold resistance and clarify their mechanisms of cold resistance,this study used annual branches of Vitis vinifera'Pinot Noir'and'Frontenac'as test materials,establishing two treatments:a control at 4℃and a low-temperature stress at ﹣20℃for 24 hours.The relative electrical conductivity(REC),soluble sugar(SS)content,and peroxidase(POD)activity were determined,and transcriptome sequencing was used to analyze the cold resistance mechanism.The results showed that under low-temperature stress,the REC,SS content,and POD activity of both varieties increased.In the transcriptome analysis,the distribution of upregulated/downregulated differentially expressed genes(601/610)in'Frontenac'was balanced,while the downregulated genes in'Pinot Noir'accounted for a higher proportion(upregulated/downregulated:348/930).GO enrichment analysis revealed that the differentially expressed genes of'Frontenac'were concentrated in phenylpropanoid metabolism,while those of'Pinot Noir'were focused on ethylene response.KEGG enrichment analysis showed that the enriched pathways of both varieties were mainly metabolic pathways,with variety-specific differences.Furthermore,MYB family transcription factors had the highest enrichment level among the differentially expressed genes of the two varieties,and the qRT-PCR verification results of 8 genes were highly consistent with the transcriptome data,confirming the reliability of the sequencing results.This study reveals the differential mechanisms of wine grape varieties differing in cold resistance in response to low-temperature stress at the physiological and molecular levels,providing a theoretical basis for cold-resistant cultivation and management of wine grapes in the Inner Mongolia region.
