基于UPLC-Q-Orbitrap高分辨质谱的现制饮料中20种合成着色剂及染料的分析方法OA
Analytical method for 20 synthetic colorants and dyes in freshly prepared beverages using UPLC-Q-Orbitrap HRMS
本研究构建了一种基于超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UPLC-Q-Orbitrap HRMS)的快速检测体系,可同步分析现制饮料中的11种合成着色剂与9种染料.采用乙腈提取-水稀释前处理技术,以10 mmol/L乙酸铵水溶液和含0.05%乙酸的乙腈作为流动相进行梯度洗脱,在C18 MG Ⅲ色谱柱上实现有效分离.检测运用电喷雾离子源正负离子切换模式,借助全扫描/数据依赖二级质谱(Full MS/dd-MS2)采集模式获取高精度质量数据.方法学验证结果显示:20种目标物在0.0038~0.12 mg/kg范围内线性良好(R²>0.9971),平均加标回收率处于72.6%~107.9%区间,精密度RSD在2.0%~8.5%范围.该方法具备操作简便、灵敏度高、结果可靠等特性,能够满足现制饮料中合成着色剂及染料的高通量筛查与定量检测需求.
A rapid detection system based on UPLC-Q-Orbitrap HRMS for simultaneous analysis of 11 synthetic colorants and 9 dyes in freshly prepared beverages was developed.A simple acetonitrile extraction-water dilution pretreatment was employed,with gradient elution performed on a C18 MG Ⅲ column using 10 mmol/L ammonium acetate aqueous solution and acetonitrile containing 0.05%acetic acid as mobile phases.Detection was carried out in positive/negative ion switching mode via electrospray ionization,and high-accuracy mass data were acquired using full scan/data-dependent MS2(Full MS/dd-MS2)acquisition.Method validation demonstrated excellent linearity(R2>0.9971)for all the 20 target analytes in the range of 0.0038~0.12 mg/kg,with average spiked recoveries of 72.6%~107.9%and precision RSDs of 2.0%~8.5%.The method was characterized by simplicity,high sensitivity,and reliability,meeting the requirements for high-throughput screening and quantitative detection of synthetic colorants and dyes in freshly prepared beverages.
