为探究党参、黄芪、茯苓复合饮料对高脂血症大鼠的降脂保肝作用及机制,将 48 只大鼠随机分为空白对照组、模型组、辛伐他汀组和党参、黄芪、茯苓复合饮料低、中、高剂量组(518、1036、2072 mg/kg),连续灌胃 8 周后,使用试剂盒测定大鼠血脂、血清转氨酶、血清中参与血脂代谢相关酶的活性和肝组织中氧化应激指标、炎症因子;Western Blot 法测定大鼠肝组织中 AMPK、wnt1/β-catenin、SIRT1 信号通路相关蛋白表达量.结果表明,与模型组相比,党参、黄芪、茯苓复合饮料能显著降低大鼠的TC、TG、AST、ALT、ACC、GPAT、FAS、MDA、IL-6、TNFα 水平(P<0.01),显著升高大鼠的 SOD、GSH-PX、HSL、LPL 水平(P<0.01),显著增加大鼠体内的 p-AMPK、WNT1、β-catenin、SIRT1、PGC-1α、Nrf2 的蛋白表达水平(P<0.01),显著降低大鼠体内的 SREBP-1C、HMGCR、C/EBP、PPARγ、NF-κB 的蛋白表达水平(P<0.01,P<0.05).说明党参、黄芪、茯苓复合饮料能够有效降血脂和减轻肝损伤,其机制与激活 AMPK、Wnt1/β-catenin、SIRT1信号通路,抑制脂肪酸合成、促进脂肪酸分解、抑制炎症反应、提高抗氧化活性相关.

To investigate the lipid-lowering and hepatoprotective effects and underlying mechanisms of the composite beverage containing Codonopsis pilosula,Astragalus membranaceus,and Poria cocos on hyperlipidemic rats,48 rats were randomly divided into the blank control group,the model group,the simvastatin group,and low-,medium-,and high-dose groups of the composite beverage(518,1036,and 2072 mg/kg,respectively).The composite beverages were gavaged to the rats daily for 8 weeks;the levels of rats blood lipids,serum transaminases,activities of enzymes involved in lipid metabolism,oxidative stress indicators,and inflammatory factors in rats liver tissue were determined using reagent kits;Western blot analysis was used to measure the expression levels of AMPK,Wnt1/β-catenin,and SIRT1 signaling pathway related proteins in rats liver tissue.Compared with the model group,the composite beverage containing Codonopsis pilosula,Astragalus membranaceus,Poria cocos significantly reduced the rats levels of TC,TG,AST,ALT,ACC,GPAT,FAS,MDA,IL-6,and TNF-α(P<0.01),and significantly increased the rats levels of SOD,GSH-PX,HSL,and LPL(P<0.01);it also significantly increased the rats protein expression levels of p-AMPK,Wnt1,β-catenin,SIRT1,PGC-1α,and Nrf2(P<0.01),and significantly decreased the rats protein expression levels of SREBP-1C,HMGCR,C/EBP,PPARγ,and NF-κB(P<0.01,P<0.05).In conclusion,the composite beverage containing Codonopsis pilosula,Astragalus membranaceus,and Poria cocos could effectively lower blood lipids and alleviate liver damage in hyperlipidemic rats.Its mechanism was associated with the activation of the AMPK,Wnt1/β-catenin,and SIRT1 signaling pathways,inhibition of fatty acid synthesis,promotion of fatty acid catabolism,suppression of inflammatory responses,and enhancement of antioxidant activity.
