为研究贝莱斯芽孢杆菌SN-1胞外多糖(Released Exopolysaccharides-2,rEPS-2)对人肝癌HepG2 细胞抑制和凋亡的影响,该文评估了 rEPS-2 对人 HepG2 细胞的抑制作用,经 rEPS-2 处理 48 h 的 HepG2 细胞,用倒置显微镜和荧光显微镜观察 rEPS-2 作用后的细胞形态,采用 MTT 法考察经 rEPS-2 处理 24 和 48 h 后对人肝癌 HepG2 细胞抑制作用,并采用流式细胞仪对处理过的 HepG2 细胞进行细胞周期分布的检测.结果表明:rEPS-2 对人肝癌 HepG2 细胞的抑制作用呈时间和剂量依赖关系,当 rEPS-2 的质量浓度为 2 mg·mL-1 时,其抑制率可达 72.35%.流式细胞仪 Annexin-V/PI 双染法分析表明,G1/S 期发生阻滞并且 G0/G1 期细胞的百分比升高,并有剂量依赖性,此外,rEPS-2 对凋亡相关活性酶 Caspase-9和Caspase-3 的活力均有所提高,经 rEPS-2 处理 48 h 后,Caspase-3 和 Caspase-9 的活性分别可达 140%和 162%.综上所述,贝莱斯芽孢杆菌 SN-1 胞外多糖对人肝癌 HepG2 细胞有明显的体外活性抑制作用,可为贝莱斯芽孢杆菌 SN-1 胞外多糖的开发和应用提供参考和数据支撑.

We investigated the impacts of exopolysaccharides isolated from Bacillus velezensis SN-1(referred to as rEPS-2)on human hepatocellular carcinoma HepG2 cells,including their influence on apoptosis.HepG2 cells were treated with rEPS-2 for 48 hours,and their morphological characteristics were observed using an inverted microscope and fluorescence microscopy.An MTT assay was employed to examine the effects of rEPS-2 treatment for 24 and 48 hours on HepG2 cell survival and proliferation.The impact of treatment on cell cycle distribution was analyzed by flow cytometry.The inhibitory effects of rEPS-2 on HepG2 cells were time and dose dependent.At an rEPS-2 concentration of 2 mg·mL-1,inhibition of cell proliferation reached 72.35%.Flow cytometry analysis using Annexin V/PI double staining indicated dose dependent induction of G1/S phase arrest,with an increased percentage of cells in G0/G1 phase.rEPS-2 also increased the activity of apoptosis-associated enzymes Caspase-9 and Caspase-3.After 48 hours of treatment with rEPS-2,Caspase-3 and Caspase-9 activities increased to 140%and 162%,respectively.In conclusion,rEPS-2 displays significant in vitro antiproliferative efficacy against HepG2 cells.These findings provide a reference and supportive data for rEPS-2 development and application.
