外源H2O2对'无核白'葡萄ROS积累的影响OA
Effects of Exogenous H2O2 on the Accumulation of ROS in'Thompson Seedless'Grapevine
为探究外源过氧化氢(H2O2)在葡萄中的分布和响应规律,本研究以'无核白'组培苗和盆栽苗为试材,采用根部培养(0~6 mmol·L-1,7 d)和叶面喷施(0~800 mmol·L-1)H2O2处理,利用氯化硝基四氮唑蓝(NBT)和二氨基联苯胺(DAB)染色法观察活性氧(ROS)实时积累,并测定过氧化氢酶(CAT)、超氧化物歧化酶(SOD)及谷胱甘肽还原酶(GR)等3种抗氧化酶活性.结果显示,根部2 mmol·L-1 H2O2处理时,叶脉H2O2没有显著积累,但是在叶缘有显著增加.叶片SOD活性在2 mmol·L-1处理时达峰值.盆栽苗叶面喷施100 mmol·L-1 H2O2处理时叶片CAT、SOD、GR活性均已显著增强.结果表明,外源H2O2不同处理方式影响叶片ROS积累规律.本研究为解析葡萄逆境应答机制提供依据.
To investigate the distribution and response mechanisms of exogenous hydrogen peroxide(H2O2)on reactive oxygen species(ROS)accumulation pattern in grapes,tissue culture seedlings and potted seedlings of'Thompson Seedless'were treated with root incubation(0-6 mmol·L-1,7 d)and foliar spraying(0-800 mmol·L-1)H2O2 applications.ROS accumulation was visualized using nitrotetrazolium blue chloride(NBT)and diaminobenzidine(DAB)staining,while activities of catalase(CAT),superoxide dismutase(SOD),and glutathione reductase(GR)were determined.Results demonstrated that H2O2 were significantly accumulated in leaf margins but not around leaf veins when roots were treated with 2 mmol·L-1 H2O2.Leaf SOD activity peaked at the 2 mmol·L-1 treatment.Leaf CAT,GR and SOD activities were all significantly enhanced in 100 mmol·L-1 pot seedling treatment group.These findings indicate that different exogenous H2O2 applications affect ROS accumulation patterns in leaves.This study provided insights and knowledge for grape stress response mechanisms.
