基于计算机技术筛选多活性蛋黄蛋白肽OA

Computer-based screening of multi-bioactive egg yolk-derived peptides

建立一种多活性整合的快速筛选体系,以提高蛋黄蛋白肽的筛选效率并降低成本.本研究在前期从蛋黄蛋白中筛选得到的DPP4 抑制肽的基础上,采用AnOxPePred、AHTpin及BIOPEP-UWM计算机工具,深度挖掘蛋黄蛋白肽的多种生物功能活性,对蛋黄蛋白肽的抗氧化、血管紧张素转换酶(ACE)抑制及α-葡萄糖苷酶(α-GC)抑制活性进行多维度评估,筛选出RYHFPEGL(RL8)和YRLRF(YF5)两种蛋黄蛋白肽;并通过体外实验对RL8 和YF5 两种蛋白肽的活性进行验证,发现RL8 和YF5 均表现出显著的多重活性.RL8[DPPH 的IC50:(2.67±0.11)mmol/L和ABTS 的IC50:(1.61±0.30)mmol/L]的自由基清除能力略优于YF5[DPPH 的IC50:(3.13±0.12)mmol/L和ABTS 的IC50:(1.94±0.13)mmol/L],其ACE和α-GC抑制活性[IC50值分别为(62.45±4.11)μmol/L和(0.67±0.07)mmol/L]也略高于YF5[ACE和α-GC抑制IC50 值分别为(78.16±2.99)μmol/L和(0.81±0.50)mmol/L].最后基于分子对接技术在分子水平上解析目标肽与靶蛋白的结合机制,发现RL8 和YF5 通过疏水作用、氢键及静电相互作用与靶酶紧密结合.该研究不仅丰富了蛋黄源生物活性肽的种类与活性数据库,也为其他来源活性肽的多活性挖掘提供参考思路.

A rapid integrated screening system was established to improve the efficiency and reduce the cost of selecting multifunctional egg yolk peptides.Building upon the DPP4-inhibitory peptides previously screened from egg yolk proteins,the computational tools AnOxPePred,AHTpin,and BIOPEP-UWM were applied to screening egg yolk peptides bioactivities.Antioxidant,angiotensin-converting enzyme(ACE)inhibitory,and α-glucosidase(α-GC)inhibitory activities of egg yolk peptides were evaluated in a multidimensional manner,and two peptides,RYHFPEGL(termed RL8)and YRLRF(termed YF5),were selected.Bioactivities of RL8 and YF5 were validated by in vitro assays,and both peptides were shown to exhibit pronounced multiactivity.Radical scavenging activity against DPPH and ABTS was slightly higher for RL8(DPPH IC50=2.67±0.11 mmol/L;ABTS IC50=1.61±0.30 mmol/L)than for YF5(DPPH IC50=3.13±0.12 mmol/L;ABTS IC50=1.94±0.13 mmol/L).ACE and αGC inhibitory potencies were also higher for RL8(ACE IC50=62.45±4.11 μmol/L;αGC IC50=0.67±0.07 mmol/L)than for YF5(ACE IC50=78.16±2.99 μmol/L;αGC IC50=0.81±0.50 mmol/L).Finally,molecular docking technology was employed to elucidate the binding mechanism between the target peptides and the target protein at the molecular level.RL8 and YF5 were found to tightly bind to the target enzyme through hydrophobic interactions,hydrogen bonds,and electrostatic interactions.This study not only enriches the database of egg yolk-derived bioactive peptides in terms of diversity and activity but also provides a reference approach for exploring multiple bioactivities of peptides from other sources.
