针对桑椹药渣中残留多糖未被充分利用的问题,优化超声辅助复合酶法提取工艺,并探究其体外抗氧化活性.以桑椹药渣为原料,通过单因素试验筛选纤维素酶与果胶酶作为复合酶,优化酶解时间、料液比(g/mL)、酶浓度、pH值、反应温度等参数,结合正交试验确定最佳提取工艺;采用苯酚-硫酸法测定多糖含量,并通过DPPH自由基和ABTS+自由基清除实验评价抗氧化活性.结果显示正交试验优化后最佳工艺为pH值 4、酶解温度 40℃、复合酶浓度 1%,多糖得率达 443.15 mg/g.体外抗氧化实验表明,桑椹药渣多糖对DPPH自由基和ABTS+自由基清除率最高分别为49.37%(10 mg/mL)和79.39%(5 mg/mL),且清除能力随浓度增加而增强.因此,超声辅助复合酶法可高效提取桑椹药渣多糖,其抗氧化活性显著,为桑椹资源综合利用及中药废弃物高值化开发提供了理论依据.

An ultrasoundassisted complex enzyme extraction process was optimized to extract residual polysaccharides from Morus alba L.(mulberry)medicinal residue,and the in vitro antioxidant activity of the obtained crude polysaccharides was investigated to support comprehensive utilization of the residue.Mulberry residue was used as the raw material,single-factor experiments were initially conducted to select the optimal enzyme combination of cellulase and pectinase,the parameters including enzymolysis time,solid-to-liquid ratio(g/mL),enzyme concentration,pH and reaction temperature were optimized.An orthogonal experimental design was then applied to determine the optimal extraction conditions.Polysaccharide content was determined by the phenol-sulfuric acid method,and antioxidant activity was evaluated by DPPH and ABTS free radical scavenging assays.The optimal conditions identified by the orthogonal test were pH 4,enzymolysis temperature 40℃and complex enzyme concentration 1%,under which a polysaccharide yield of 443.15 mg/g was achieved.The in vitro antioxidant assays demonstrated that the mulberry polysaccharides exhibited significant radical scavenging capacity.The maximum scavenging rates were 49.37%(at 10 mg/mL)for DPPH free radicals and 79.39%(at 5 mg/mL)for ABTS free radicals,and the scavenging capacity was observed increasing with polysaccharide concentration.It was concluded that the ultrasound-assisted complex enzyme method was an efficient and effective process for extracting polysaccharides from mulberry residue.The extracted polysaccharides exhibited significant antioxidant activity,providing a theoretical foundation for the comprehensive utilization of mulberry resources and high-value development of this traditional Chinese medicine waste.
