复合酶辅助提取瓶尔小草多糖工艺优化及其体外抗氧化活性OA
Optimization of composite enzyme-assisted extraction process of polysaccharides from Ophioglossum vulgatum and their in vitro antioxidant activity
以瓶尔小草为原材料,在前期单因素试验的基础上,选取液料比、提取温度、提取时间、复合酶用量(纤维素酶∶木瓜蛋白酶质量比 1∶1)四个因素为自变量,以多糖提取得率为评价指标,运用四因素三水平的响应面法优化瓶尔小草多糖提取工艺;同时,通过体外抗氧化实验,测定瓶尔小草多糖对DPPH自由基和ABTS+自由基的清除能力,以评价其抗氧化活性.经优化得到的最佳酶解提取工艺为:液料比 66∶1(mL/g)、提取温度 52℃、复合酶用量 0.94%、提取时间 90 min.在此条件下,瓶尔小草多糖的得率为 13.84%,与预测值相符.瓶尔小草多糖具有较好的抗氧化活性,其DPPH自由基和ABTS+自由基的IC50 值分别为 40.25、48.07 μg/mL.
The enzymatic extraction of polysaccharides from Ophioglossum vulgatum was optimized,and the extract's in vitro antioxidant activity was evaluated.Based on the previous single factor tests,the response surface methodology(RSM)was applied.The independent variables were the liquid-to-solid ratio,extraction temperature,extraction time,and the concentration of the compound enzyme(cellulase∶papain at a mass ratio of 1∶1).Using polysaccharide extraction yield as the evaluation criterion,a four-factor,three-level response surface method was employed to optimize the extraction process for the polysaccharides from Ophioglossum vulgatum.The optimized enzymatic extraction process was determined as a liquid-to-solid ratio of 66∶1(mL/g),extraction temperature of 52℃,composite enzyme dosage of 0.94%and extraction time of 90 min.Under the above conditions,the yield of the polysaccharide was 13.84%,consistent with the predicted value.Furthermore,the extracted polysaccharide demonstrated good antioxidant potential,its scavenging ability against DPPH free radicals and ABTS free radicals were strong.The IC50 values were 40.25 and 48.07 μg/mL,respectively.These findings highlight that the polysaccharides from Ophioglossum vulgatum are a promising source of natural antioxidants.
