益生菌安全性评价研究进展OA
Research progress on safety evaluation of probiotics
随着大健康产业的发展,益生菌越来越多地进入人们的日常饮食生活,对其进行菌株水平的安全性评价对人类健康具有重要意义.该研究基于国内外专家研究成果及益生菌安全性相关指导文件,系统梳理了益生菌安全性评价的研究进展,归纳了安全性评价的标准流程,并对未来评价方法的发展趋势进行了展望.益生菌的安全性评价流程包括国内外安全性评价资料分析、菌株的分类鉴定、体外评价和体内评价四部分.国内外安全性评价资料分析是指对菌种国内、国外使用历史及安全性(包括但不限于其致病性和产毒能力报告、临床试验资料、科技文献或综述等)进行综合分析.菌株分类鉴定最有效的方法是全基因组测序,通过对益生菌的毒力基因、不良特性相关基因以及耐药基因进行分析来评估菌株安全性.体外评价包括对益生菌的溶血活性、产生物胺的量、抗生素的耐药性以及产D-乳酸的量进行检测.体内评价主要是基于动物模型的毒理学评价.该文为选择益生菌安全性评价方法以及替代毒理学模型提供新的思路和理论资料.
With the development of the health industry,probiotics are increasingly becoming part of people's daily dietary lives.Con-ducting safety evaluations of probiotics at the strain level is of significant importance for human health.This study systematically reviews the research progress on the safety evaluation of probiotics based on the research achievements of experts both domestically and internationally,as well as related guidance documents on probiotic safety.It summarizes the standard procedures for safety evaluation and provides an out-look on the future development trends of evaluation methods.The safety evaluation process for probiotics comprises four components,inclu-ding analysis of domestic and international safety evaluation data,strain classification and identification,in vitro assessments,and in vivo evaluations.Analysis of domestic and foreign safety evaluation data refers to a comprehensive analysis of the domestic and foreign use histo-ry and safety of bacterial species(including but not limited to their pathogenicity and toxicity-producing ability reports,clinical trial data,scientific and technological literature or reviews,etc.).The most effective method for strain classification and identification is whole ge-nome sequencing,which assesses strain safety by analyzing virulence genes,genes related to undesirable characteristics,and antibiotic re-sistance genes in probiotics.In vitro evaluation includes testing for hemolytic activity,biogenic amine levels,antibiotic resistance,and D-lactic acid production.In vivo evaluation is primarily based on toxicological evaluation using animal models.This paper provides new ideas and theoretical references for selecting probiotic safety evaluation methods and alternative toxicological models.
