金钱草及其易混淆品的特异性PCR鉴别OA
Identification of Lysimachiae Herba and Its Adulterants by Specific PCR
目的:建立一种快速鉴别金钱草及其易混淆品的特异性聚合酶链式反应(PCR)方法.方法:依据 ITS基因序列中金钱草(基原过路黄)及其混淆品的差异位点,设计过路黄特异性鉴别引物;提取各样品 DNA 后建立PCR 反应体系,考察影响 PCR 反应的主要因素,并对该方法进行适用性验证,扩增产物通过电泳进行检测.结果:当退火温度为 60℃,循环次数为 30 次时,过路黄样品均在 174 bp 处有单一清晰的目标条带,在同样条件下,其混淆品均无此条带.结论:建立的特异性 PCR 方法可准确且较快速地鉴别金钱草及其混淆品,为金钱草药材的物种真实性鉴定及质量评价提供了技术支持.
Objective:To establish a specific polymerase chain reaction(PCR)method for the rapid identification of Lysimachiae Herba and its adulterants.Methods:Based on the differential loci in the ITS gene sequences between Lysimachiae Herba(original plant Lysimachia christinae)and its adulterants,specific identification primers for Lysimachia christinae were designed.DNA was extracted from various samples,and a PCR reaction system was established.The main factors affecting the PCR reaction were investigated,and the method was validated for applicability.The amplified products were detected by electrophoresis.Results:When the annealing tempera-ture was set at 60℃and the cycle number was 30,all samples of Lysimachia christinae showed a single,clear target band at 174 bp,while no such band appeared in the adulterant samples under the same conditions.Conclusion:The specific PCR method established in this study can rapidly and accurately identify Lysimachiae Herba and its adulterants,providing technical support for the authenticity and quality evaluation of Lysimachiae Herba medicinal material.
