Expression and prognostic significance of the APRO family genes in head and neck squamous cell carcinoma

Objective To systematically analyze the gene expression profiles of the anti-proliferative protein(APRO)family in head and neck squamous cell carcinoma(HNSC),clarify their correlations with patient prognosis and immune cell infiltration, and evaluate their potential value as prognostic biomarkers and therapeutic targets for HNSC.Methods Based on The Cancer Genome Atlas(TCGA)public database, the UALCAN platform was used to analyze the differential expression of the APRO family between HNSC tissues and normal tissues,as well as their correlations with clinicopathological parameters. The Kaplan-Meier Plotter online survival analysis platform was applied to assess the relationships between differentially expressed genes and patients' prognosis. The TIMER2.0 immune microenvironment analysis platform was utilized to evaluate the correlations of differentially expressed genes with immune cell infiltration in HNSC.The Metascape platform was adopted to perform GO and KEGG functional enrichment analyses on the co-expressed genes of differentially expressed APRO family genes. The cBioPortal platform was employed to analyze the genetic alteration characteristics and co-occurrence pattern of the APRO family. Results Compared with normal tissues,TOB1 and BTG2 in the APRO family were downregulated, whereas TOB2,BTG1 and BTG3 were upregulated in HNSC tissues(P<0.05).The expression levels of the above five genes were correlated with the clinical staging of HNSC(P<0.05).Low expressions of BTG1 and BTG3 were significantly associated with poor prognosis in HNSC patients(P<0.05).TOB1,TOB2,BTG1,BTG2 and BTG3 were all closely correlated with the infiltration of immune cells such as neutrophils(P<0.05).Co-expressed genes of the APRO family were mainly enriched in transcriptional co-regulator activity and lysine degradation pathway. The APRO family exhibited a certain mutation rate in HNSC,dominated by gene amplification and missense mutation, and showed significant co-occurrence patterns. Conclusion The APRO family exhibits a specific expression pattern in HNSC.Its expression levels are closely associated with patients' clinical staging, prognosis and immune cell infiltration. Moreover, characteristic genetic alterations are observed in this family, indicating that it can serve as a potential prognostic biomarker and gene therapy target for HNSC.
