血清miR-124-3p和miR-214-3p对年龄相关性白内障的诊断价值OA
Diagnostic value of serum miR-124-3p and miR-214-3p in age-related cataract
目的 探讨血清miR-124-3p和miR-214-3p水平对年龄相关性白内障(ARC)的诊断价值.方法 选取2022年1月至2023年12月在西宁爱尔眼科医院就诊的98例ARC患者作为研究组.另选取同期在在西宁爱尔眼科医院体检的95例健康体检者作为对照组.采用实时荧光定量聚合酶链反应检测所有研究对象血清miR-124-3p、miR-214-3p水平,采用酶联免疫吸附试验检测所有研究对象血清超氧化物歧化酶(SOD)、谷胱氨酸过氧化物酶(GSH-Px)、丙二醛(MDA)水平.收集所有研究对象基线资料.采用Pearson相关分析ARC患者血清miR-124-3p、miR-214-3p水平与SOD、MDA、GSH-Px水平的相关性.采用多因素Lo-gistic 回归分析发生ARC的影响因素.绘制受试者工作特征(ROC)曲线分析血清miR-124-3p、miR-214-3p单独及二者联合检测对发生ARC的诊断价值.结果 研究组血清MDA水平高于对照组,血清SOD、GSH-Px、miR-124-3p、miR-214-3p水平均低于对照组,差异均有统计学意义(P＜0.05).Pearson相关分析结果显示,ARC患者血清miR-124-3p水平与血清SOD、GSH-Px水平均呈正相关(P＜0.05),与血清MDA水平呈负相关(P＜0.05);血清miR-214-3p水平与SOD、GSH-Px水平均呈正相关(P＜0.05),与血清MDA水平呈负相关(P＜0.05).多因素Logistic回归分析结果显示,血清SOD、GSH-Px、miR-124-3p、miR-214-3p水平升高均为发生ARC的保护因素(P＜0.05),血清MDA水平升高为发生ARC的危险因素(P＜0.05).血清miR-124-3p、miR-214-3p单独及二者联合检测诊断ARC发生的曲线下面积(AUC)分别为0.756、0.770、0.837,二者联合诊断的 AUC 大于 miR-124-3p、miR-214-3p 单独诊断的 AUC(Z=1.782、1.525,P＜0.05).结论 ARC 患者血清miR-124-3p和miR-214-3p水平均降低,且参与ARC的发生、发展,二者联合对ARC的发生具有一定的诊断价值.
Objective To investigate the diagnostic value of serum miR-124-3p and miR-214-3p levels in age-related cataract(ARC).Methods A total of 98 ARC patients who were treated in Xining Aier Eye Hospi-tal from January 2022 to December 2023 were selected as the study group.Another 95 healthy people who un-derwent physical examination in Xining Aier Eye Hospital during the same period were selected as the control group.The serum levels of miR-124-3p and miR-214-3p in all subjects were detected by real-time fluorescence quantitative polymerase chain reaction.The serum levels of superoxide dismutase(SOD),glutathione peroxi-dase(GSH-Px)and malondialdehyde(MDA)were detected by enzyme-linked immunosorbent assay in all subjects.Baseline data of all subjects were collected.Pearson correlation was used to analyze the correlation between serum miR-124-3p,miR-214-3p and SOD,MDA,GSH-Px levels in ARC patients.Multivariate Logis-tic regression was used to analyze the influencing factors of ARC.The receiver operating characteristic(ROC)curve was drawn to analyze the diagnostic value of serum miR-124-3p,miR-214-3p alone and combined detec-tion for ARC.Results The serum MDA level of the study group was higher than that of the control group,and the serum SOD,GSH-Px,miR-124-3p and miR-214-3p levels were lower than those of the control group,and the differences were statistically significant(P＜0.05).Pearson correlation analysis showed that serum miR-124-3p level in ARC patients was positively correlated with serum SOD and GSH-Px levels(P＜0.05),and negatively correlated with serum MDA level(P＜0.05).The level of serum miR-214-3p was positively correlated with the levels of SOD and GSH-Px(P＜0.05),and negatively correlated with the level of serum MDA(P＜0.05).The results of multivariate Logistic regression analysis showed that the increase of serum SOD,GSH-Px,miR-124-3p and miR-214-3p levels were protective factors for ARC(P＜0.05),and the in-crease of serum MDA level was a risk factor for ARC(P＜0.05).The area under the curve(AUC)of serum miR-124-3p,miR-214-3p alone and combined detection in the diagnosis of ARC was 0.756,0.770 and 0.837,respectively.The AUC of the combined detection in the diagnosis of miR-124-3p and miR-214-3p was greater than that of miR-124-3p or miR-214-3p alone(Z=1.782,1.525,P＜0.05).Conclusion The levels of serum miR-124-3p and miR-214-3p in ARC patients are decreased,and they are involved in the occurrence and devel-opment of ARC.The combination of miR-124-3p and miR-214-3p has certain diagnostic value for ARC.
