基于Notch信号通路探讨金龙定喘合剂调节免疫失衡治疗哮喘的作用机制OA
Exploring Mechanism of Jinlong Dingchuan Mixture(金龙定喘合剂)Regulating Immune Imbalance in Treating Asthma Based on Notch Signaling Pathway
目的 基于Notch信号通路探讨金龙定喘合剂调节支气管哮喘大鼠辅助性T细胞17(Th17)/调节性T细胞(Treg)平衡的作用机制.方法 50只SD大鼠随机分为对照组、模型组、金龙定喘合剂组、Notch激动剂组及金龙定喘合剂+Notch激动剂组,每组10只.注射和吸入卵白蛋白(OVA)方式制备大鼠哮喘模型,各药物组大鼠灌胃/腹腔注射相应药物,1次/d,连续14 d.检测大鼠静息每分钟通气量(VE)、呼气峰流速(PEF),以评价肺功能;HE和Masson染色观察肺组织病理损伤与纤维化情况;检测支气管肺泡灌洗液(BALF)中总细胞计数、嗜酸性粒细胞、中性粒细胞,淋巴细胞数量;流式细胞仪测定外周血中Th17、Treg比例;ELISA测定BALF中白细胞介素(IL)-10、IL-17、转化生长因子-β(TGF-β)水平,Western blot检测肺组织中Notch1及发状分裂相关增强子1(Hes1)蛋白的表达.结果 金龙定喘合剂组大鼠肺功能显著改善,肺组织病理损伤与纤维化明显减轻;BALF中炎症细胞数量显著减少;Th17/Treg比值显著降低,IL-10、TGF-β水平显著升高,IL-17水平显著降低;Notch1与Hes1蛋白表达水平显著降低;而Notch激动剂Jagged1可部分逆转金龙定喘合剂对哮喘大鼠的上述治疗作用.结论 金龙定喘合剂能够改善支气管哮喘大鼠Th17/Treg平衡,抑制炎症反应,改善肺功能,其作用机制可能与抑制Notch信号通路有关.
Objective To explore the mechanism of Jinlong Dingchuan Mixture(金龙定喘合剂)in regulating the balance of helper T cell 17(Th17)/regulatory T cell(Treg)in the bronchial asthma mice based on Notch signaling pathway.Methods Fifty C57BL/6 mice were randomly divided into control group(control),model group(model),Jinlong Dingchuan Mixture group(JLDC),Notch agonist group(Jagged1)and Jinlong Dingchuan Mixture+Notch agonist group(JLDC+Jagged1)(n=10).The asthma model was prepared by injection and inhalation of ovalbumin(OVA).The mice in each drug group were given the corre-sponding drug by gavage/intraperitoneally once a day for 14 consecutive days.Resting minute ventilation(VE)and peak expira-tory flow rate(PEF)were measured to evaluate the lung function.HE and Masson staining methods were used to observe the pathological injury and fibrosis of lung tissue.The total cell count,eosinophils,neutrophils and lymphocytes were detected in bronchoalveolar lavage fluid(BALF).Flow cytometry was used to determine the proportion of helper T cells 17(Th17)and regu-latory T cells(Treg)in peripheral blood.The levels of interleukin(IL)-10,IL-17 and transforming growth factor-β(TGF-β)in bronchoalveolar lavage fluid(BALF)were measured,and the expressions of Notch1 and Hes1 protein in lung tissue were detected.Results The lung function of JLDC group was significantly improved,and the pathological injury and fibrosis of the lung tissue were significantly alleviated.The number of inflammatory cells in BALF decreased significantly.Th17/Treg ratio was sig-nificantly decreased,the levels of IL-10 and TGF-β were significantly increased,and IL-17 levels were significantly de-creased.The expression levels of Notch1 and Hes1 were significantly decreased.Notch agonist Jagged1 can partially reverse the above therapeutic effects of Jinlong Dingchuan Mixture on the asthmatic mice.Conclusion Jinlong Dingchuan Mixture can im-prove Th17/Treg balance,inhibit inflammatory response and improve lung function in the mice with tracheal asthma,and its mechanism may be related to inhibiting Notch signaling pathway.
