目的 基于超高效液相色谱四极杆飞行时间质谱(UPLC/Q-TOF MS)研究经方天麻钩藤饮对抽动障碍模型小鼠的干预及脑组织中纹状体代谢物的影响,以探究经方天麻钩藤饮治疗抽动障碍的保护机制.方法 取60只ICR小鼠(雄性),分为6组,每组10只,分别为对照组、模型组、西药组、天麻钩藤饮高剂量组、天麻钩藤饮中剂量组、天麻钩藤饮低剂量组.通过亚氨基二丙腈(Iminodipropionitrile,IDPN)构建小鼠抽动障碍模型,综合小鼠行为学变化和组织病理,结合小鼠脑组织多巴胺(Dopamine,DA)、5-羟色胺(5-Hydroxytryptamine,5-HT)与去甲肾上腺素(Norepinephrine,NE)的含量,初步评价经方天麻钩藤饮对抽动障碍模型小鼠的治疗效果.利用UPLC/Q-TOF MS技术检测各组小鼠脑组织中的纹状体代谢物,以正交偏最小二判别分析进行多元统计分析模式识别,筛选差异代谢物,并借助MetaboAnalyst5.0数据库富集代谢通路.结果 经方天麻钩藤饮可以显著改善抽动障碍模型小鼠的刻板行为评分,降低肺指数,5-HT检测量下调,DA、NE检测量上升.相比于对照组,模型组小鼠脑组织5-HT检测量显著升高(P<0.05),DA和NE检测量显著降低(P<0.05);相较于模型组,天麻钩藤饮高、中、低剂量组小鼠脑组织5-HT均下降,DA、NE均上升(P<0.05);进一步对比可见,天麻钩藤饮高剂量组小鼠脑组织中5-HT含量较其中、低剂量组更低,DA、NE含量则更高(P<0.05).代谢组学结果显示,与经方天麻钩藤饮调控相关的差异代谢物共有50余个,包含甘油磷脂、亚油酸、花生四烯酸等物质.结论 从代谢组学角度分析了经方天麻钩藤饮治疗抽动障碍的作用机制,为经方天麻钩藤饮治疗抽动性障碍症研究及临床应用奠定了基础.

Objective This study aimed to investigate the mechanism of Tianma Gouteng Decoction(天麻钩藤饮)in the treat-ment of tic disorders by examining its intervention effects on tic disorder model mice and its impact on striatal metabolites in brain tissue using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC/Q-TOF MS).Methods Sixty male ICR mice were chosen and randomly allocated into 6 groups with 10 mice in each,specifically the con-trol group,the model group,the western medicine group,as well as the Tianma Gouteng Decoction high,medium and low dose groups.A tic disorder model in mice was constructed via intraperitoneal injection of iminodipropionitrile(IDPN).The therapeutic effect of Tianma Gouteng Decoction on tic disorder model mice was preliminarily evaluated by comprehensively considering chan-ges in mouse behavior,histopathology,as well as the levels of dopamine(DA),5-hydroxytryptamine(5-HT)and norepineph-rine(NE)in the brain tissue of mice.UPLC/Q-TOF MS was used to detect striatal metabolites in the brain tissue of the mice in the control group,the model group,the western medicine group and Tianma Gouteng Decoction high,medium and low dose groups.Orthogonal partial least squares-discriminant analysis was adopted for multivariate statistical analysis and pattern recog-nition,aiming to screen out differential metabolites.Metabolic pathway enrichment was performed using the MetaboAnalyst 5.0 database.Results Tianma Gouteng Decoction could significantly improve the stereotyped behavior scores of tic disorder model mice,reduce the lung index,lower the level of 5-HT and raise the levels of DA and NE.In particular,when compared with those in the control group,the level of 5-HT in the brain tissue of the mice in the model group was significantly higher(P<0.05),whereas the levels of DA and NE were significantly reduced(P<0.05).In comparison with the model group,the 5-HT levels in the brain tissue of the mice in the Tianma Gouteng Decoction high,medium and low dose groups were significantly lowered,while the levels of DA and NE were significantly elevated(P<0.05).Further comparison showed that the content of 5-HT in the brain tissue of the mice in the Tianma Gouteng Decoction high-dose group was lower than that in the Tianma Gouteng Decoc-tion medium and low dose groups,while the contents of DA and NE were higher(P<0.05).Metabolomics results revealed that there were more than 50 differential metabolites related to the regulation of Tianma Gouteng Decoction,including glycerophospho-lipids,linoleic acid and arachidonic acid.Conclusion From a metabolomics standpoint,this research explored the therapeutic mechanism of Tianma Gouteng Decoction in addressing tic disorders,laying a foundation for research and clinical application of Tianma Gouteng Decoction in the treatment of tic disorders.
