党参及杂交群体的表型变异与党参炔苷含量分析OA
Analysis of Phenotypic Variation and Lobetyolin Content in Codonopsis pilosula and Its Hybrid Populations
渭党和纹党是甘肃地区主栽的两种中药党参.依托远缘杂交育种路径,以两种党参各35份及两者杂交产生的127份F1群体为材料,在室内水培条件下,对根茎叶的13个形态性状与党参根的炔苷含量进行系统测定及相关性分析.结果表明,两亲本在形态性状均值上差异不显著,但渭党党参炔苷含量极显著高于纹党;纹党群体平均遗传多样性指数以及平均变异系数高于渭党群体;F1群体平均遗传多样性指数(1.56)高于双亲,而平均变异系数(47.57%)低于双亲,表现为性状分布更均匀但波动幅度降低;主成分与聚类分析显示F1群体形成新的性状组合结构;多数地上部性状表现显著中亲优势和超亲优势,呈正向超亲分离;党参炔苷含量在亲本中与形态性状相关性较弱,但在F1群体中与根粗、分根数及根长呈极显著正相关.研究揭示了纹党与渭党之间形态差异有限,杂交后代表型变异丰富,为党参种质创新与优良性状选择提供了理论依据.
Codonopsis pilosula(Franch.)Nannf.and Codonopsis pilosula var.modesta(Nannf.)L.T.Shen are two major medicinal germplasm widely cultivated in Gansu province,China.Based on distant hybridization breeding,35 accessions of each parent along with 127 F1 hybrid progenies derived from their cross were utilized as experimental materials.Thirteen morphological traits of roots,stems and leaves,along with the lobetyolin content in roots,were systematically measured and analyzed under indoor hydroponic conditions.The results demonstrated that the two parents exhibited no significant differences in the mean values of morphological traits,whereas C.pilosula showed a highly significantly higher lobetyolin content than C.pilosula var.modesta.However,the C.pilosula var.modesta population displayed a higher average genetic diversity index and average coefficient of variation than C.pilosula population.The F1 population achieved the highest average genetic diversity index(1.56)compared to both parents,while its average coefficient of variation was lower than that of the parents,indicating a more stable phenotypic structure with reduced fluctuation amplitude.Principal component and hierarchical cluster analyses revealed that the F1 population formed a novel trait combination structure.Most above-ground traits exhibited significant mid-parent and transgressive heterosis,presenting positive transgressive segregation.Lobetyolin content was weakly correlated with morphological traits within the parental populations.Crucially,within the F1 population,lobetyolin content showed highly significant positive correlations with root diameter,number of lateral roots,and root length(P<0.01).This study reveals that although the morphological differences between the two parental varieties are limited,their hybrid progenies display abundant phenotypic variation and structural reconfiguration between quality and morphological traits,providing a theoretical basis for germplasm innovation and indirect selection of superior agronomic traits in C.pilosula
