水蛭素抗肿瘤作用研究进展OA
Research progress on the anti-tumor effect of hirudin
水蛭素是传统中药水蛭的主要活性成分,具有抗凝、抗炎、抗纤维化、抗肿瘤等多重药理作用.其可通过调控细胞因子和凝血相关靶基因的表达,影响PI3K/AKT、ERK/MAPK等信号通路活性,抑制神经胶质瘤、肺癌、肝癌、生殖系统肿瘤及膀胱癌等多种恶性肿瘤的进展.其抗肿瘤机制既包括直接抑制肿瘤细胞增殖、迁移并诱导凋亡,也涵盖通过抗凝作用改善肿瘤微环境、抑制血管生成及上皮-间质转化(EMT)等间接途径.本文结合肿瘤发生发展与治疗现状,系统综述水蛭素在不同肿瘤中的作用及其分子机制,并对当前研究的局限与未来临床应用方向进行探讨,以期为推动其进一步研究与临床转化提供理论依据.
Hirudin is the main active ingredient of hirudo in traditional Chinese medicine.It has anti-coagulation,anti-inflammation,anti-fibrosis and anti-tumor effects.Studies have confirmed that hirudin inhibits the progression of various malignant tumors,including glioblastoma,lung cancer,hepatocellular carcinoma,reproductive system tumors,and bladder cancer,by regulating the expression of cytokines and coagulation-related target genes and influencing the activity of signal-ing pathways such as PI3K/AKT and ERK/MAPK.Its anti-tumor mechanisms involve both direct effects such as inhibiting tumor cell proliferation and migration while inducing apoptosis,and indirect pathways mediated by its anticoagulant proper-ties,including improving the tumor microenvironment,suppressing angiogenesis,and inhibiting epithelial-mesenchymal transition(EMT).This review systematically summarizes the role and molecular mechanisms of hirudin in different tumors in the context of tumor development and current treatment approaches.It also discusses existing research limitations and fu-ture directions for clinical application,aiming to provide a theoretical foundation for advancing further research and clinical translation of hirudin.
