温和灸足三里对再生障碍性贫血小鼠造血功能及Notch信号通路相关因子表达的影响OA
Effects of mild moxibustion at Zusanli(ST36)on hematopoietic function and expression of Notch signaling pathway-related factors in aplastic anemia mice
目的:观察温和灸足三里对再生障碍性贫血(AA)小鼠造血功能及Notch信号通路关键因子表达的影响,探索温和灸治疗AA的可能机制.方法:共28只雄性C57BL/6J小鼠,随机选取10只为正常组,其余18只采用联合注射环磷酰胺(CTX)和环孢霉素(Cs)的方法制备小鼠AA模型.模型验证成功后,造模小鼠随机分为模型组和艾灸组,每组8只.正常组小鼠自然喂养,仅做与艾灸组相同的固定及抓取.造模后艾灸组小鼠接受温和灸单侧足三里干预,每日1次,每次15 min.两侧穴位交替使用.连续干预60 d.观察各组小鼠骨髓细胞增生情况和骨髓造血组织结构;采用流式细胞术检测骨髓CD34+细胞比例;采用免疫印迹法和实时荧光定量聚合酶链反应检测骨髓CD34+细胞中Jagged经典Notch配体1(Jagged1)、Notch受体1(Notch1)、Delta样经典Notch配体4(DLL4)、毛状分裂相关增强子1(Hes1)蛋白和mRNA的表达水平.结果:与正常组相比,模型组小鼠骨髓造血细胞计数和造血组织比例显著降低(P<0.01);与模型组相比,艾灸组小鼠骨髓造血细胞计数和造血组织比例显著增加(P<0.01,P<0.05).与正常组相比,模型组小鼠骨髓中CD34+细胞比例显著下降(P<0.01);与模型组相比,艾灸组小鼠骨髓中CD34+细胞比例显著上升(P<0.01).与正常组相比,模型组小鼠骨髓CD34+细胞Notch1、Jagged1、DLL4、Hes1蛋白和mRNA表达显著下降(P<0.01);与模型组相比,艾灸组小鼠骨髓CD34+细胞上述蛋白和mRNA表达显著上升(P<0.01).结论:温和灸足三里对AA小鼠的造血功能具有改善作用,其机制可能与Notch信号通路有关.
Objective:To observe the effects of mild moxibustion at Zusanli(ST36)on hematopoietic function and expression of Notch signaling pathway key factors in aplastic anemia(AA)mice,thereby exploring the potential mechanism of mild moxibustion in AA treatment. Methods:Ten mice were randomly selected as the normal control group from 28 male C57BL/6J mice,and the other 18 were injected with cyclophosphamide(CTX)and cyclosporine(Cs)to prepare the AA model.Successfully validated model mice were randomly divided into a model group and a moxibustion group,with 8 mice in each group.Mice in the normal control group were fed routinely,with same fixation and grasping performed as in the moxibustion group.After modeling,mice in the moxibustion group received mild moxibustion at unilateral Zusanli(ST36),once daily for 15 min each time.Bilateral Zusanli(ST36)were used alternately with consecutive moxibustion for 60 d.The proliferation of bone marrow cells and the structure of bone marrow hematopoietic tissue were observed in each group.The proportion of CD34+cells in bone marrow was detected by flow cytometry.Detection of protein and mRNA expression levels of Jagged canonical Notch ligand 1(Jagged1),Notch receptor 1(Notch1),delta-like ligand 4(DLL4),and Hes family BHLH transcription factor 1(Hes1)in bone marrow CD34+cells was performed by Western blotting and real-time fluorescence quantitative polymerase chain reaction. Results:Compared with the normal control group,hematopoietic cell numbers and hematopoietic tissue proportions in mouse bone marrow in the model group were significantly decreased(P<0.01).Compared with the model group,hematopoietic cell numbers and hematopoietic tissue proportions in mouse bone marrow in the moxibustion group were significantly increased(P<0.01,P<0.05).Compared with the normal control group,the CD34+cell proportion in mouse bone marrow in the model group was decreased significantly(P<0.01).Compared with the model group,CD34+cell proportion in mouse bone marrow in the moxibustion group was increased significantly(P<0.01).Compared with the normal control group,the protein and mRNA expression levels of Notch1,Jagged1,DLL4,and Hes1 in mouse bone marrow CD34+cells in the model group were significantly decreased(P<0.01).Compared with the model group,the above protein and mRNA expression levels in mouse bone marrow CD34+cells in the moxibustion group were significantly increased(P<0.01). Conclusion:Mild moxibustion at Zusanli(ST36)improves hematopoietic function of AA mice,and the mechanism may be related to the Notch signaling pathway.
