紫草素调控PTEN/AKT/TFEB信号通路改善心肌梗死大鼠心室重构OA

目的 探讨紫草素(SK)通过调控磷酸酶和紧张素同源物(PTEN)/蛋白激酶 B(protein kinase B,AKT)/转录因子 EB(TFEB)信号通路改善心肌梗死(MI)心室重构的作用机制.方法 将SD大鼠随机分成Sham组、MI组、L-SK组、H-SK组和H-SK+B-pV组,每组10只.采用心脏超声、组织学染色、酶联免疫吸附试验(ELISA)及Western blotting分别检测各组大鼠心功能、心肌组织病理变化、血清炎症因子水平及相关蛋白表达.结果 与Sham组相比,MI组大鼠心肌纤维化、炎症细胞浸润显著增加,心功能指标恶化;与MI组相比,紫草素干预组(L-SK组、H-SK组)心肌损伤减轻,纤维化程度降低,心功能显著改善,且呈剂量依赖性,同时PTEN/AKT/TFEB信号通路相关蛋白及自噬相关蛋白表达发生显著变化;H-SK+BpV组则呈现相反趋势.结论 紫草素可能通过调控PTEN/AKT/TFEB信号通路抑制炎症反应,激活心肌细胞自噬,从而改善MI大鼠心功能,减轻心肌损伤和纤维化.

Objective To investigate the mechanism by which shikonin(SK)improves ventricular remodeling in myocardial infarc-tion(MI)by modulating the phosphatase and tensin homolog(PTEN)/protein kinase B(AKT)/transcription factor EB(TFEB)signaling pathway.Methods SD rats were randomly divided into Sham,MI,L-SK,H-SK,and H-SK+BpV groups(n=10 each).Cardiac function,myocardial histopathology,serum inflammatory factor levels,and protein expressions were assessed via echocardiography,histological staining,ELISA,and Western blotting.Results Compared with Sham group,MI group exhibited increased myocardial fibrosis,inflamma-tory infiltration,and deteriorated cardiac function.Compared with the MI group,SK-treated groups(L-SK,H-SK)showed reduced myocar-dial injury and fibrosis,and improved cardiac function in a dose-dependent manner,along with significant changes in PTEN/AKT/TFEB and autophagy-related proteins.The H-SK+BpV group demonstrated opposite trends.Conclusion SK may improve cardiac function and attenuate myocardial injury and fibrosis in MI rats by modulating the PTEN/AKT/TFEB signaling pathway,suppressing inflammation,and activating cardiomyocyte autophagy.
