EGCG调节小脑浦肯野细胞兴奋性改善VPA诱导小鼠行为缺陷OA
EGCG modulates cerebellar Purkinje cell excitability to ameliorate VPA-induced behavioral deficits in mice
目的 本研究旨在探究腹腔注射表没食子儿茶素没食子酸酯(EGCG)对丙戊酸钠(VPA)诱导孤独症(ASD)小鼠小脑浦肯野细胞兴奋性和抑制性突触活性的改变及其对小鼠孤独症样刻板行为、运动协调与平衡障碍的干预效果.方法 构建VPA诱导ASD小鼠模型,分别腹腔注射25 mg/kg和50 mg/kg EGCG,通过行为学方法(埋球实验、自我梳理实验、加速转棒实验、平衡木实验)观察正常对照(CON)组、VPA组、VPA+25 mg/kg EGCG组和VPA+50 mg/kg EGCG组小鼠在重复刻板行为、运动协调与平衡障碍的差异;通过电生理记录观察各组小鼠小脑浦肯野细胞兴奋性和抑制性突触活性的改变.结果 埋球实验和自我梳理实验中50 mg/kg EGCG干预可显著减轻VPA诱导ASD小鼠重复刻板行为(P<0.01);加速转棒实验和平衡木实验中50 mg/kg EGCG干预可显著改善VPA诱导ASD小鼠运动协调和平衡能力(P<0.01);全细胞膜片钳模式记录浦肯野细胞电生理变化,通过50 mg/kg EGCG干预可明显改善VPA诱导ASD小鼠小脑浦肯野细胞兴奋性降低(P<0.05)和抑制性突触活性降低(P<0.05,P<0.01),但不影响其兴奋性突触活性(P>0.05).结论 EGCG能够剂量依赖性地调节VPA诱导ASD小鼠小脑浦肯野细胞兴奋性和抑制性突触活性的改变,从而改善VPA诱导ASD小鼠孤独症样刻板行为、运动协调与平衡障碍.
Objective To investigate the effects of epigallocatechin-3-gallate(EGCG)administration on cerebellar Purkinje cell excitability and inhibitory synaptic activity in valproic acid(VPA)-induced autism spectrum disorder(ASD)mice,as well as its effects on autism-like stereotyped behaviors and motor coordination/balance deficits.Methods A VPA-induced ASD mouse model was established via intraperitoneal injections of EGCG(25 mg/kg and 50 mg/kg).Behavioral tests,including marble burying,self-grooming,accelerating rotarod,and balance beam tests,were used to assess repetitive stereotyped behaviors and motor coordination/balance among four groups:normal control,VPA,VPA+25 mg/kg EGCG,and VPA+50 mg/kg EGCG.Electrophysiological recordings were used to investigate alterations in both excitatory and inhibitory synaptic activity of cerebellar Purkinje cells across these groups.Results In the marble burying and self-grooming tests,50 mg/kg EGCG significantly alleviated repetitive stereotyped behaviors in VPA-induced ASD mice(P<0.01).In the accelerated rotarod and balance beam tests,50 mg/kg EGCG markedly improved motor coordination and balance abilities in VPA-induced ASD mice(P<0.01).Whole-cell patch-clamp recordings demonstrated that 50 mg/kg EGCG effectively ameliorated VPA-induced reductions in both cerebellar Purkinje cell excitability(P<0.05)and inhibitory synaptic activity(P<0.05,P<0.01),without affecting excitatory synaptic activity(P>0.05).Conclusions EGCG dose-dependently modulated the altered excitability and inhibitory synaptic activity of cerebellar Purkinje cells,thereby ameliorating autism-like stereotyped behaviors and motor coordination and balance impairments in VPA-induced ASD mice.
