储存时间对茅苍术挥发性成分分析及其抗炎活性的影响OA

为探讨储存时间对茅苍术质量的影响,本试验采用气相色谱-质谱联用技术(GC-MS)系统分析不同储存年限(0、1、2 年)茅苍术的挥发性成分动态变化,并通过脂多糖(LPS)诱导的小鼠单核巨噬细胞 RAW 264.7 炎症模型评价其抗炎活性.结果表明:茅苍术醇、β-桉叶醇、苍术素等主要挥发性成分在储存初期(0 年)呈现最高含量(分别占总挥发性成分的 40.71%、19.37%、6.39%),随储存时间延长呈下降趋势.细胞试验显示,各储存组挥发性成分在 50 μg/mL 浓度下均未表现细胞毒性(细胞存活率>90%),对 LPS 诱导的 NO 释放具有抑制活性(抑制率29.08%~31.75%),其中储存 1 年茅苍术挥发性成分表现出相对最佳抗炎活性.本研究揭示了茅苍术挥发性成分的储存稳定性与其抗炎活性的非线性关系,为制定中药材科学仓储标准提供了关键试验依据.

The study aimed to investigate the impact of storage duration on the quality of Atractylodes lancea(AL).GC-MS was employed to systematically analyze the dynamic changes in volatile components of AL stored for different periods(0,1year,and 2 years),while LPS-induced RAW 264.7 murine monocyte-macrophage inflammatory models were utilized to evaluate anti-inflammatory activity.The results demonstrated that characteristic volatile components,including atractylodin,hinesol,and β-eudesmol,exhibited the highest initial content during fresh storage(0 year,accounting for 40.71%,19.37%and 6.39%of total volatile components,respectively),with a gradual decline observed over extended storage periods.Cellular experiments revealed no cytotoxicity for volatile components across all storage groups at 50 μg/mL(cell viability>90%),with consistent inhibition of LPS-induced NO release(inhibition rate:29.08%~31.75%).Notably,the 1-year-stored AL displayed a relatively optimal anti-inflammatory trend.This study elucidate the nonlinear correlation between the storage stability of volatile components in AL and their anti-inflammatory efficacy,providing critical experimental evidence for establishing scientific storage standards of traditional Chinese medicinal materials.
