Effects of pulsatilla extract on growth performance,serum antioxidant indices and rumen fermentation parameters of mutton sheep

文章旨在探究不同添加水平的白头翁提取物(PE)对肉羊生长性能、血清抗氧化指标及瘤胃发酵参数的影响,试验选取 180 只初始体重 25 kg 左右的湖羊公羔,随机分为 5 个处理组,各处理组设 6 个重复栏,每栏 6 只.CK 饲喂基础日粮,4 个添加组(PE1、PE2、PE3、PE4)分别添加 200、400、800 和 1600 mg/kg 的 PE.预试期 5 d,正试期 90 d.结果表明:(1)生长性能方面,PE2和PE3的末重(FW)分别较CK显著提高5.84%和8.96%(P<0.05),平均日增重(ADG)分别显著提高 8.69%和 13.37%(P<0.05),料重比(F/G)分别显著降低 6.67%和 9.33%(P<0.05),其中 PE3 的改善效果显著优于 PE2(P<0.05);PE4 的 FW 较 CK 显著降低 6.12%(P<0.05),ADG 显著降低 17.01%(P<0.05),F/G 显著提高 17.00%(P<0.05).(2)血清抗氧化及血液生化指标方面,与 CK 相比,PE2 和 PE3 的总抗氧化能力(T-AOC)、抑制羟自由基能力(IHFRC)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)及超氧化物歧化酶(SOD)活性均显著提高(P<0.05),丙二醛(MDA)含量明显下降(P<0.05),且 PE3 显著优于 PE2(P<0.05);PE1、PE2 和PE3 组的丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、血尿素氮(BUN)和肌酐(CREA)与 CK 相比均无显著差异(P>0.05);PE4 各抗氧化指标显著劣于 CK(P<0.05),ALT、AST、BUN 和 CREA 均显著高于 CK(P<0.05).(3)瘤胃发酵参数方面,PE2 和 PE3 的 MCP、乙酸、丙酸含量及总挥发性脂肪酸(TVFA)均显著高于 CK(P<0.05),氨态氮(NH3-N)和 pH 显著低于 CK(P<0.05),PE3 的瘤胃发酵参数显著优于 PE2(P<0.05);PE4 各指标显著劣于 CK(P<0.05).综合各项指标,在本试验条件下,PE 在肉羊日粮中的适宜添加量为 400~800 mg/kg,其中 800 mg/kg 添加量的改善效果最佳;超过 1600 mg/kg 的添加量会对肉羊产生显著不利影响.

This study investigated the effects of different supplemental levels of Pulsatilla extract(PE)on growth performance,serum antioxidant indices and rumen fermentation parameters of mutton sheep,and to determine the optimal addition amount of PE in the diet of mutton sheep.A total of 180 Hu sheep male lambs with an initial body weight of approximately 25 kg were divided into 5 groups.CK was fed a basal diet,and the four experimental groups(PE1,PE2,PE3 and PE4)were fed the basal diet supplemented with 200,400,800 and 1600 mg/kg Pulsatilla extract,respectively.The results showed that:1)PE1 showed no significant difference from the CK(P>0.05).Compared with CK,FW of PE2 and PE3 were significantly enhanced by 5.84%and 8.96%(P<0.05),ADG by 8.69%and 13.37%(P<0.05),and F/G were significantly reduced by 6.67%and 9.33%(P<0.05),respectively,with PE3 showing significantly better effects than PE2(P<0.05).In contrast,FW and ADG of PE4 were significantly reduced by 6.12%and 17.01%compared with CK(P<0.05),while F/G was significantly increased by 17.00%(P<0.05).2)Regarding serum antioxidant indices,the total antioxidant capacity,inhibition of hydroxyl free radical capacity,glutathione peroxidase,catalase and superoxide dismutase activities in PE2 and PE3 were significantly enhanced(P<0.05),while MDA was significantly lower than that in CK(P<0.05),and PE3 showed significantly better results than PE2(P<0.05).No significant differences were observed in ALT,AST,BUN or CREA among CK,PE1,PE2 and PE3 groups(P>0.05).All antioxidant indices in PE4 were significantly inferior to those of CK(P<0.05),while ALT,AST,BUN and CREA in PE4 were significantly higher than those in CK(P<0.05).3)Regarding rumen fermentation parameters,the microbial protein and total volatile fatty acid of PE2 and PE3 were significantly higher than those in CK(P<0.05),while the ammonia nitrogen content was significantly lower than that in CK(P<0.05),and PE3 showed significantly better results than the PE2(P<0.05).All rumen fermentation parameters in PE4 were significantly inferior to those of CK(P<0.05).Based on comprehensive evaluation of all indices,the optimal addition amount of PE is 400-800 mg/kg,with the 800 mg/kg addition showing the best improvement.An addition amount exceeding 1600 mg/kg exerts significant adverse effects on mutton sheep.
