针对养殖端促生长类抗生素全面禁用的行业现状,本试验旨在筛选高效拮抗大肠杆菌、沙门氏菌的益生菌,为绿色无抗养殖提供菌种资源.从健康肉鸡肠道内容物分离菌株,通过双层平板初筛、牛津杯法复筛获得目标菌株,结合菌落形态、革兰氏染色、16S rDNA 测序完成菌种鉴定,并系统探究菌株接种量、活性组分对抑菌效果的影响.结果显示,共分离得到 6 株具抑菌活性的肠道菌,其中 R2、R5 抑菌性能最优;形态学观察结合序列同源比对证实,R2 为革兰氏阳性短杆状芽孢杆菌,R5 为革兰氏阳性球形乳酸球菌(乳酸菌大类).牛津杯复筛中 R5 对两种致病菌净抑菌圈直径分别为 7.65 mm、7.40 mm,抑菌效果优于 R2.接种量试验表明,5%~25%接种范围内,R2 抑菌效果无显著差异(P>0.05);R5 抑制大肠杆菌、沙门氏菌的最佳接种量分别为 15%、20%.R5 抑菌活性物质主要分泌于发酵上清液,上清液处理组 OD600 显著低于菌体沉淀与全菌液组(P<0.05),其上清抑菌圈可达 10.10 mm、7.55 mm.综上所述,本研究筛选得到一株抑菌活性较好的鸡源乳酸菌 R5,可为替抗型益生菌饲料添加剂的开发提供试验支撑.

Given the ban on growth-promoting antibiotics in livestock feeds,this study aimed to screen probiotics with strong antagonistic activity against Escherichia coli and Salmonella for green antibiotic-free breeding.Strains were isolated from the intestinal contents of healthy broilers,followed by primary screening with a double-layer plate method and secondary screening via the Oxford cup assay.The strains were identified by colony morphology,Gram staining and 16S rDNA sequencing,and the effects of inoculation amount and active components on antibacterial activity were investigated.The results showed that six strains with bacteriostatic activity were isolated,among which strains R2 and R5 showed the optimal antibacterial performance.Combined with morphological observation and sequence homology alignment,strain R2 was identified as a Gram-positive short rod-shaped Bacillus,and strain R5 was identified as a Gram-positive spherical Lactococcus(a genus of lactic acid bacteria).In the Oxford cup re-screening test,the net inhibition zone diameters of strain R5 against E.coli and Salmonella were 7.65 mm and 7.40 mm,respectively,which were superior to those of strain R2.The inoculation test showed that the antibacterial effect of R2 had no significant difference within the inoculation range of 5%~25%(P>0.05);the optimal inoculation amounts of R5 for inhibiting E.coli and Salmonella were 15%and 20%,respectively.The antibacterial substances of R5 were mainly distributed in the fermentation supernatant.The OD600 of the supernatant group was significantly lower than the precipitation and whole bacterial suspension groups(P<0.05),and the inhibition zone diameters of the supernatant against two pathogens reached 10.10 mm and 7.55 mm,respectively.This study screened a chicken-derived Lactococcus strain R5 with excellent bacteriostatic activity,which provides experimental support for the development of probiotic additives replacing antibiotics.
