基于多分子标记的唐松草属发育与物种鉴定OA
Phylogenetic Analysis and Species Identification of Thalictrum Plants Based on Molecular Markers
DNA 条形码技术作为物种鉴定的重要工具,近年来在植物分类学和中药材鉴定领域展现出显著优势.本研究以唐松草属 6 个物种为研究对象,结合 ITS、rbcL、matK、ycf1 四种分子标记,通过 PCR 扩增、Sanger 测序、BLAST 比对及系统发育分析,探究其系统发育关系与物种鉴定准确性.结果显示,所有样品均成功扩增出目标序列,其中 ITS 和 ycf1 标记在物种区分中表现最优,支持率高达 100%.系统发育树表明,6 个样品均归属于唐松草亚属,其中 T1 利用分子手段确定为贝加尔唐松草,T5 与瓣蕊唐松草高度关联.研究验证了 ITS、rbcl、matK、ycf1分子标记应用的可靠性,为唐松草属植物的分类修订和中药材鉴定提供了分子证据.
DNA barcoding technology has emerged as a significant tool for species identification and has shown re-markable advantages in the fields of plant taxonomy and traditional Chinese medicine identification in recent years.This study takes six species of the genus Thalictrum as the research objects,combines four molecular markers-ITS,rbcL,matK,and ycf1-and investigates their phylogenetic relationships and the accuracy of species identification through PCR amplification,Sanger sequencing,BLAST alignment,and phylogenetic analysis.The results show that all samples were successfully amplified with the target sequences,among which the ITS and ycf1 markers performed optimally in species differentiation,with a support rate of up to 100%.The phylogenetic tree indicates that all six samples belong to the subgenus Thalictrum(Thalictrum subg.Thalictrum),where T1 was determined to be Thal-ictrum baicalense using molecular methods,and T5 is highly related to Thalictrum petaloideum.This study verifies the reliability of the combined application of multiple molecular markers and provides molecular evidence for the tax-onomic revision of Thalictrum plants and the identification of traditional Chinese medicinal materials.
