英文文献中天然黄酮化合物与阿尔茨海默病动物试验的系统评价OA
Systematic Evaluation of Natural Flavonoids and Animal Experiments of Alzheimer Disease in English Literature
目的 总结天然黄酮类化合物对于阿尔茨海默病(AD)动物模型认知功能、病理生理现象影响的证据.方法 系统检索PubMed、Web of Science、Embase英文数据库的文献,纳入AD动物模型研究,提取认知行为学指标(如水迷宫、Y迷宫、避暗实验)、病理指标(Aβ负荷、tau磷酸化、神经炎症、氧化应激等)及给药方案,并通过SYRCLE工具评估偏倚风险,采用叙述性综合分析方法总结研究结果.结果 共纳入 28 项动物研究(>1200 只动物).多数黄酮类化合物显著改善AD模型的认知功能,降低Aβ沉积和tau蛋白磷酸化水平,并减轻神经炎症和氧化应激.其中,木犀草素(n=6)、川陈皮素(n=3)、7,8-二羟基黄酮(n=5)作用证据较多.部分研究结果存在差异,如 1 项研究未观察到 7,8-二羟基黄酮改善认知功能,1 项川陈皮素研究对Aβ沉积无显著作用.结论 天然黄酮类化合物在改善认知功能、减轻Aβ沉积、抑制tau蛋白磷酸化,以及降低神经炎症和氧化应激方面具有明确的神经保护作用,但不同研究在动物模型、给药剂量和干预时长等方面存在异质性,且大部分研究的偏倚风险较高,但仍需通过标准化的长期高质量动物及临床研究来进一步验证其疗效与安全性.
Objective To summarize the evidence of the effects of natural flavonoids on cognitive function and pathophysiological phenomena in Alzheimer Disease(AD)animal models.Methods Literatures in PubMed,Web of Science and Embase databases were systematically searched.AD animal model studies were included.Cognitive behavioral indicators(such as water maze,Y maze,dark avoidance test),pathological indicators(A β load,tau phosphorylation,neuroinflammation,oxidative stress,etc.)and dosage regimens were extracted.The risk of bias was evaluated by SYRCLE tool,and the results were summarized by narrative comprehensive analysis method.Results A total of 28 animal studies(>1200 animals)were included.Most flavonoids significantly improved the cognitive function of AD models,reduced Aβ deposition and tau protein phosphorylation levels,and alleviated neuroinflammation and oxidative stress.Among them,luteolin(n=6),nobiletin(n=3),and 7,8-dihydroxyflavone(n=5)had more evidence of action.There were differences in the results of some studies.For example,7,8-dihydroxyflavone was not observed to improve cognitive function in one study,and nobiletin had no significant effect on Aβ deposition in one study.Conclusion Natural flavonoids have definite neuroprotective effects in improving cognitive function,reducing Aβ deposition,inhibiting tau protein phosphorylation,and reducing neuroinflammation and oxidative stress.However,different studies have heterogeneity in animal models,dosage and duration of intervention,and most studies have a high risk of bias.However,standardized long-term high-quality animal and clinical studies are still needed to further verify its efficacy and safety.
