基于生物信息学和CRISPR基因敲除筛选技术探索锌指蛋白28在肝细胞癌中的表达及临床意义OA
Expression and Clinical Significance of Zinc Finger Protein 28 in Hepatocellular Carcinoma Based on Bioinformatics and CRISPR Gene Knockout Screening Technology
目的 探讨锌指蛋白 28(ZNF28)在肝细胞癌(HCC)的表达及分子机制.方法 通过TCGA数据库获得ZNF28 在HCC及泛癌中的表达数据及相关临床特征参数,比较ZNF28 在癌组织和正常组织中的表达差异及预后的关系.结合CRISPR敲除筛选和CCLE数据库,分析ZNF28 基因对HCC细胞增殖的影响并评估其敲除效应.使用R软件、maftools包分析突变基因,采用基因富集分析探讨ZNF28 相关的信号通路,借助TIMER、CIBERSORT研究免疫浸润.结果 ZNF28 在HCC中的表达高于正常组织,且其高表达与患者较差预后相关(P<0.05).CRISPR敲除ZNF28 后,13 种HCC细胞系的生长和增殖速率降低.ZNF28 在大多数肿瘤组织中的表达高于正常组织,并与多种肿瘤患者的生存预后相关(P<0.05).突变分析显示,TP53 基因突变频率最高,ZNF28 可能通过调控补体和凝血级联反应、代谢通路、神经活性配体-受体相互作用等途径在HCC中发挥重要作用.在ZNF28 高表达的HCC样本中,调节性T细胞、γδT细胞、M0/M1 巨噬细胞和活化肥大细胞等免疫细胞呈现激活状态.结论 ZNF28 作为HCC诊断潜在分子标志物,可能通过影响多种信号通路参与HCC的发生发展.
Objective To investigate the expression and molecular mechanism of zinc finger protein 28(ZNF28)in hepatocellular carcinoma(HCC).Methods The expression data and related clinical characteristic parameters of ZNF28 in HCC and pan-cancerous tissues were obtained from TCGA database,and the relationship between the expression difference of ZNF28 in cancer tissues and normal tissues and the prognosis was compared.Combined with CRISPR knockout screening and CCLE database,the effect of ZNF28 gene on the proliferation of HCC cells was analyzed and its knockout effect was evaluated.R software and maftools package were used to analyze mutant genes,gene enrichment analysis was used to explore ZNF28-related signaling pathways,and TIMER and CIBERSORT were used to study immune infiltration.Results The expression level of ZNF28 in HCC was higher than that in normal tissues,and its high expression was associated with poor prognosis(P<0.05).After ZNF28 knockout by CRISPR,the growth and proliferation rates of 13 HCC cell lines were decreased.The expression level of ZNF28 in most tumor tissues was higher than that in normal tissues,and was associated with the survival and prognosis of patients with various tumors(P<0.05).Mutation analysis showed that the mutation frequency of TP53 gene was the highest,and ZNF28 might play an important role in HCC by regulating complement and coagulation cascades,metabolic pathways,and neuroactive ligand-receptor interactions.In HCC samples with high ZNF28 expression,immune cells such as regulatory T cells,γδT cells,M0/M1 macrophages and activated mast cells were activated.Conclusion A As a potential molecular marker for HCC diagnosis,ZNF28 may be involved in the occurrence and development of HCC by affecting multiple signaling pathways.
