目的:优化栀黄消肿凝胶贴膏(Zhihuang Xiaozhong gel plaster,ZGP)处方,评价其体外释放及透皮性能,并进行初步药效学考察.方法:以聚丙烯酸钠 NP-700、甘羟铝、聚乙烯吡咯烷酮(PVP K-30)、酒石酸、甘油、水为影响因素,以感官评价、初黏力、持黏力及剥离强度为评价指标,D-最优混料设计优化ZGP基质配比;以栀子苷为指标,考察ZGP经皮渗透性及体外释药性,并从不同质量分数氮酮中筛选最佳促渗剂;建立大鼠急性软组织损伤模型,以苏木素-伊红(HE)染色观察肌肉组织形态学变化,ELISA检测白细胞介素-1β(IL-1β)、前列腺素2(PGE2)水平,初步探讨其改善软组织损伤的作用.结果:ZGP的最佳处方为聚丙烯酸钠 NP-700 8.61%、甘羟铝0.16%、PVP K-30 4.84%、酒石酸0.16%、甘油45.91%、水24.19%、浸膏16.13%;栀子苷24h的累积释放率为(66.96±4.81)%,1%氮酮的促渗效果最佳.体内药效学实验表明,与模型组比较,ZGP能够显著改善肌肉组织损伤,降低PGE2、IL-1β水平,差异具有显著统计学意义(P<0.001).结论:优化后的ZGP制备工艺稳定可行,体外释药性和经皮渗透性能良好,显示出良好的抗炎效果,为栀黄消肿方外用新剂型的开发提供了实验依据.

Objective:To optimize the formulation of Zhihuang Xiaozhong gel plaster(ZGP)and evaluate its in vitro re-lease,transdermal properties,and preliminary pharmacodynamic effects.Methods:The matrix composition of ZGP was optimized using a D-optimal mixture design with sodium polyacrylate NP-700,dihydroxyaluminum aminoacetate,poly-vinylpyrrolidone(PVP K-30),tartaric acid,glycerol,and water as influencing factors.Evaluation indicators included sensory properties,initial adhesion,holding power,and peel strength.Using geniposide as the marker compound,the in vitro release and transdermal permeation of ZGP were investigated,and the optimal concentration of azone as a penetra-tion enhancer was screened from different mass fractions.A rat model of acute soft tissue injury was established.Mor-phological changes in muscle tissue were observed via hematoxylin-eosin(HE)staining,and the levels of interleukin-1β(IL-1β)and prostaglandin E2(PGE2)were detected by ELISA to preliminarily explore its therapeutic effect on soft tis-sue injury.Results:The optimal formulation of ZGP was:sodium polyacrylate NP-700 8.61%,dihydroxyaluminum aminoacetate 0.16%,PVP K-30 4.84%,tartaric acid 0.16%,glycerol 45.91%,water 24.19%,and extract 16.13%.The cumulative release rate of geniposide at 24 hours was(66.96±4.81)%,and 1%azone showed the best penetra-tion-enhancing effect.Pharmacodynamic experiments demonstrated that,compared with the model group,ZGP signifi-cantly improved muscle tissue damage and reduced the levels of PGE2 and IL-1β(P<0.001).Conclusion:The opti-mized ZGP exhibited a stable and feasible preparation process,favorable in vitro drug release and transdermal perme-ation properties,and significant anti-inflammatory effects,providing an experimental basis for developing a novel topical dosage form of the Zhihuang Xiaozhong formula.
