内障丸对早发性卵巢功能不全的作用效果分析OA
Analysis of the Effect of Neizhang Pill on Premature Ovarian Insufficiency
目的:探讨内障丸对早发性卵巢功能不全(POI)的作用效果.方法:取48只SPF级健康雌性SD大鼠,采用头颈三角区皮下注射 D-半乳糖200mg/kg,连续干预42d,建立早发性卵巢功能不全(POI)模型,按体质量将SD大鼠随机分为空白对照组(NC)、模型对照组(MC)、阳性对照组(PC)、内障丸低剂量组(L)、内障丸中剂量组(M)、内障丸高剂量组(H).分别运用 HE染色观察大鼠卵巢组织病理状态,免疫组化法观察大鼠卵巢组织雌激素受体(ER)、孕激素受体(PR)表达水平,采用ELISA法观察雌激素(E2)、卵泡刺激素(FSH)、促黄体生成素(LH)、白细胞介素-6(IL-6)含量.结果:HE结果显示内障丸可使早发性卵巢功能不全大鼠模型卵巢成熟卵泡增多,闭锁卵泡减少,黄体增多而体积大,卵泡周围颗粒细胞形态正常.免疫组化结果表明内障丸治疗可以增加早发性卵巢功能不全大鼠模型卵巢ER和PR表达水平.ELISA法表明内障丸可减少早发性卵巢功能不全大鼠模型卵巢FSH、LH、IL-6含量,提高E2 含量.结论:内障丸干预POI效果明显,能够有效促进早发性卵巢功能不全大鼠模型生殖系统水平的恢复,为临床提供参考.
Objective:To explore the effect of Neizhang Pill on Premature Ovarian Insufficiency(POI).Methods:Forty SPF grade healthy female SD rats were subcutaneously injected with 200 mg/kg D-galactose in the head neck triangle area for 42 consecutive days to establish a premature ovarian insufficiency(POI)model.The SD rats were randomly divided into a blank control group(NC),a model control group(MC),a positive control group(PC),a low-dose Neizhang Pill group(L),a medium dose Neizhang Pill group(M),and a high-dose Neizhang Pill group(H)according to body weight.HE staining was used to observe the pathological status of rat ovarian tissue,immunohistochemistry was used to observe the expression levels of estrogen receptor(ER)and progesterone receptor(PR)in rat ovarian tis-sue,and ELISA was used to observe the contents of estradiol(E2),follicle stimulating hormone(FSH),luteinizing hormone(LH),and interleukin-6(IL-6).Results:HE results showed that Neizhang Pill can increase the number of mature follicles,reduce the number of blocked follicles,increase the volume of corpus luteum,and normalize the mor-phology of granulosa cells around follicles in a rat model of early-onset ovarian dysfunction.The immunohistochemical results showed that treatment with Neizhang Pill can increase the expression levels of ER and PR in the ovaries of a rat model of early-onset ovarian dysfunction.The ELISA method showed that Neizhang Pill can reduce the levels of FSH,LH,and IL-6 in the ovaries of rats with early-onset ovarian dysfunction,and increase the level of E2.Conclusion:The intervention effect of Neizhang Pill on POI is significant,and it can effectively promote the recovery of the reproductive system level in a rat model of early-onset ovarian dysfunction,providing support for clinical promotion.
