基于代谢组学探讨电针预处理对脑缺血损伤大鼠脑皮质代谢物的影响OA
Exploring the Effect of Electroacupuncture Pretreatment on Cortical Metabolites in Rats with Cerebral Ischemic Injury Based on Metabolomics
目的:基于非靶向代谢组学技术研究电针预处理对脑缺血损伤大鼠脑皮质代谢物的影响,探讨电针预处理诱导脑保护的分子机制.方法:将30只雄性SD大鼠按随机数字表法分为假手术组、模型组、电针预处理组,每组10只.电针预处理组电针"百会"、双侧"内关"和"三阴交",疏密波,频率2/15Hz,20min/次,连续治疗5d,于末次电针后2h采用线栓法制备大脑中动脉闭塞模型.假手术组、模型组仅给予抓取束缚,不进行电针干预.造模24h后分别采用 Garcia JH 评分评估神经行为功能;苏木素伊红(HE)染色观察大鼠脑组织病理改变;非靶向代谢组学技术分析脑皮质代谢物的改变.结果:与假手术组相比,模型组大鼠Garcia JH评分降低(P<0.05),HE染色示缺血半暗带皮层脑组织疏松,水肿严重;与模型组相比,电针预处理组大鼠 Garcia JH 评分升高(P<0.05),缺血半暗带皮层脑组织层次尚清晰,水肿减轻;代谢组学结果显示,模型组和电针预处理组共鉴定出差异代谢物16个(8个上调,8个下调),通路富集为核黄素代谢、谷胱甘肽代谢通路.结论:电针预处理可以减轻脑缺血损伤大鼠神经缺损程度,改善脑组织病理损伤,发挥脑保护作用,其具体机制与调控核黄素代谢、谷胱甘肽代谢等通路有关.
Objective:To utilize non-targeted metabolomics techniques so as to investigate the effects of electroacupunc-ture pretreatment on the metabolite profile of the cerebral cortex in rats with ischemic brain injury,and to clarify the molecular pathways of the neuroprotective effect of electroacupuncture pretreatment.Methods:For the experimental design,thirty male Sprague-Dawley rats were allocated into three groups using a randomization process:sham operation group,model group,and electroacupuncture preconditioning group(n=10 per group).In the electroacupuncture pre-conditioning group,rats underwent electroacupuncture stimulation at acupoints"Baihui(GV20)",bilateral"Neiguan(PC06)",and"Sanyinjiao(SP6)".The stimulation parameters consisted of dense-disperse waveforms at a frequency of 2/15 Hz,administered for 20 minutes per session over five consecutive days.Following the final electroacupuncture ses-sion,two hours later,the middle cerebral artery occlusion model was induced via intraluminal suture embolization.The sham operation group and model group were only subjected to grasping and binding without electroacupuncture inter-vention.At 24h after modeling,neurological function was evaluated by Garcia JH score,pathological changes of brain tissue were observed by hematoxylin-eosin(HE)staining,and changes in cerebral cortical metabolites were analyzed by non-targeted metabolomics technology.Results:When compared to the sham operation group,the model group ex-hibited a significantly reduced Garcia JH score(P<0.05).Histopathological analysis via HE staining revealed disor-ganized brain tissue with marked edema in the ischemic penumbra cortex.In contrast,the electroacupuncture precondi-tioning group demonstrated a statistically elevated Garcia JH score compared to the model group(P<0.05),with HE staining showing well-defined cortical layers and decreased edema in the ischemic penumbra region.Metabolomic profi-ling identified 16 differentially expressed metabolites between the model and electroacupuncture preconditioning groups,including 8 up-regulated and 8 down-regulated metabolites.Pathway analysis showed that electroacupuncture preconditioning exerted brain protection mainly by regulating riboflavin metabolism and glutathione metabolism path-ways.Conclusion:Electroacupuncture preconditioning can alleviate neurological deficit and improve pathological dam-age of brain tissue in rats with cerebral ischemic injury,and its brain protective mechanism is related to regulating ribo-flavin metabolism,glutathione metabolism,and other pathways.
