大柴胡汤对2型糖尿病小鼠的疗效及肠道菌群调节作用OA
Therapeutic Effects of Dachaihu Decoction on Type 2 Diabetes Mellitus Mice and Its Regulation of the Gut Microbiome
目的:运用16S rRNA测序技术研究大柴胡汤对2型糖尿病(T2DM)小鼠的疗效及对肠道菌群的调节作用.方法:30只6周龄雄性C57BL/6J小鼠通过高脂饮食联合链脲佐菌素(STZ)腹腔注射复制T2DM 小鼠模型.造模成功后,采用随机数字表法将小鼠均分为模型组、二甲双胍组、大柴胡汤组,另设同期雄性C57BL/6J小鼠为正常组,每组10只.大柴胡汤组与二甲双胍组予以相应药物灌胃,正常组与模型组予以等体积生理盐水灌胃,每日1次,连续8周.给药期间监测各组小鼠体质量及空腹血糖(FBG).末次给药后,进行口服糖耐量试验(OGTT)与胰岛素耐量试验(ITT),然后麻醉取材.全自动生化仪检测各组小鼠肝功能及血脂水平;苏木精-伊红染色法(HE)与油红 O染色法观察肝脏组织病理变化及脂质沉积;16S rRNA测序方法观察各组小鼠肠道微生态菌落及结构的变化.结果:与模型组比较,大柴胡汤干预显著降低了 T2DM 小鼠体质量、FBG水平(P<0.01),并显著改善了糖耐量和胰岛素抵抗(P<0.01).同时,大柴胡汤能有效纠正血脂代谢紊乱,显著降低血清TC、TG、LDL-C水平(P<0.01),升高 HDL-C水平(P<0.01),降低 ALT、AST活性(P<0.01).HE与油红 O染色结果显示大柴胡汤显著改善了肝脏病理变化与脂质沉积.16S rRNA测序结果显示,大柴胡汤显著增加了T2DM 小鼠肠道菌群的α多样性,并逆转了其整体菌群结构的改变.在门水平上,大柴胡汤显著降低了厚壁菌门/拟杆菌门(Firmicutes/Bacteroidota)的比值;LEfSe分析表明,大柴胡汤组富集了以拟杆菌门(Bacteroidota)为主的标志性菌群.PICRUSt2功能预测揭示大柴胡汤可下调异常活跃的碳水化合物代谢(Carbohydrate metabolism)与膜转运(Membrane transport)通路,上调受抑制的氨基酸代谢(Amino acid metabolism)、聚糖生物合成与代谢(Glycan Biosynthesis and metabolism)以及能量代谢(Energy metabolism)等通路.相关性分析进一步揭示,大柴胡汤调控的菌群变化与改善的代谢表型显著相关.结论:大柴胡汤能有效改善 T2DM 小鼠的糖脂代谢紊乱、胰岛素抵抗及肝脏脂肪变性,其作用机制可能与调节肠道菌群结构、恢复菌群生态平衡及功能密切相关.
Objective:To investigate the therapeutic efficacy of Dachaihu Decoction in a mouse model of type 2 diabetes mellitus(T2DM)and its regulatory effects on the gut microbiota using 16S rRNA sequencing.Methods:The T2DM model was established in thirty 6-week-old male C57BL/6J mice by inducing them with a high-fat diet and intraperito-neal injections of streptozotocin(STZ).Post-modeling,the mice were randomly allocated into a model group,a Dachaihu decoction group,and a metformin group,with 10 mice per group.A normal control group consisting of age-matched male C57BL/6J mice was also included.The Dachaihu Decoction and metformin groups received their respec-tive treatments via daily gavage for 8 weeks,while the normal and model groups received an equivalent volume of nor-mal saline.Body weight and fasting blood glucose(FBG)were monitored throughout the treatment period.At the end of the 8 weeks,an oral glucose tolerance test(OGTT)and an insulin tolerance test(ITT)were conducted,after which the mice were anesthetized for sample collection.Liver function and serum lipid profiles were analyzed using an auto-mated biochemical analyzer.Histopathological changes and lipid accumulation in the liver were assessed through He-matoxylin and Eosin(H&E)and Oil Red O staining.Alterations in the gut microbial community and structure were e-valuated using 16S rRNA sequencing.Results:Compared to the model group,treatment with Dachaihu Decoction led to a significant reduction in body weight and FBG levels(P<0.01)and a marked improvement in glucose tolerance and insulin resistance(P<0.01).Dachaihu decoction also effectively ameliorated dyslipidemia,sig-nificantly lowering serum total cholesterol(TC),triglycerides(TG),and low-density lipoprotein cholesterol(LDL-C)(P<0.01),while increasing high-density lipoprotein cholesterol(HDL-C)(P<0.01).Further-more,Dachaihu decoction decreased the activity of alanine aminotransferase(ALT)and aspartate aminotrans-ferase(AST)(P<0.01).H&E and Oil Red O staining results showed that Dachaihu decoction significantly improved liver pathological changes and lipid deposition.16S rRNA sequencing revealed that Dachaihu Decoc-tion administration significantly increased the α-diversity of the gut microbiota and reversed the overall micro-bial dysbiosis observed in the model group.At the phylum level,Dachaihu Decoction decreased the Firmi-cutes/Bacteroidota ratio.Linear discriminant analysis Effect Size(LEfSe)identified a significant enrichment of Bacteroidota as a signature phylum in the Dachaihu decoction group.PICRUSt2 functional prediction indi-cated that Dachaihu Decoction downregulated hyperactivated carbohydrate metabolism and membrane trans-port pathways,while upregulating suppressed pathways related to amino acid metabolism,glycan biosynthesis and metabolism,and energy metabolism.Correlation analysis confirmed a strong association between the mi-crobial shifts induced by Dachaihu Decoction and the improved metabolic phenotypes.Conclusion:Dachaihu Decoction effectively ameliorates metabolic disorders,insulin resistance,and hepatic steatosis in T2DM mice.The underlying mechanism is likely closely associated with the modulation of gut microbiota composition,res-toration of its ecological balance,and normalization of its functional pathways.
