千层纸素A对胃癌SGC-7901细胞的影响及其调控铁死亡机制研究OA

探究千层纸素A对胃癌SGC-7901细胞的作用及机制.采用细胞活力试剂盒、Hoechst 33258荧光探针、细胞划痕、Transwell小室实验、活性氧(reactive oxygen species,ROS)荧光探针、JC-1荧光探针、亚铁离子(Fe2+)荧光探针、网络药理学、蛋白质免疫印迹实验探究千层纸素A对胃癌SGC-7901细胞活力、凋亡、迁移、侵袭及铁死亡的作用及其调控铁死亡机制.结果发现,千层纸素A能够抑制SGC-7901细胞活力、划痕愈合、迁移和侵袭,以及诱导细胞凋亡、ROS、线粒体膜电位极化和Fe2+水平.此外,千层纸素A能够降低细胞表皮生长因子受体(epidermal growth factor receptor,EGFR)蛋白表达.EGFR 激动剂 NSC 228155 能够逆转千层纸素A诱导的细胞Fe2+水平升高和细胞活力降低.结果表明,千层纸素A具有明确的抗胃癌SGC-7901细胞作用,并通过抑制EGFR诱导细胞铁死亡.

The study investigated the effects and mechanisms of oroxylin A on gastric cancer SGC-7901 cells.The methods employed included cell viability assay kit,Hoechst 33258 fluorescent probe,cell scratch assay,Transwell assays,reactive oxygen species(ROS)fluorescent probe,JC-1 fluorescent probe,ferrous ion(Fe2+)fluorescent probe,network pharmacology,and Western blot to explore the impact of oroxylin A on cell viability,apoptosis,migration,invasion,and ferroptosis,as well as its regulatory mechanisms of ferroptosis in SGC-7901 cells.The results revealed that oroxylin A inhibited SGC-7901 cell viability,scratch healing,migration,and invasion,as well as induced apoptosis,ROS accumulation,mitochondrial membrane potential depolarization,and Fe2+levels.Additionally,oroxylin A reduced the expression of the epidermal growth factor receptor(EGFR)protein in cells.The EGFR agonist NSC 228155 can reverse the increase in Fe2+levels and the decrease in cell viability induced by oroxylin A.The results indicate that oroxylin A exhibits a clear anticancer effect on gastric cancer SGC-7901 cells and induces cellular ferroptosis through the inhibition of EGFR.
