基于细胞的卷烟烟气中主要甜感物质的甜度值测定OA

为克服目前烟气甜度测定方法的局限性,实现卷烟感官感受的客观量化表征,通过构建高表达人源甜味受体T1R2/T1R3的HEK293细胞系,并使用钙离子成像技术实时监测甜感物质诱导的细胞内钙离子信号变化,实现了甜味响应的定量化表征.在此基础上,系统测定了包括糠醛和呋喃酮等10种已报道的烟气中代表性甜感物质单体的味觉甜度值,进一步考察了总粒相物(TPM)对这些甜感物质甜度表现的影响.结果表明:①多数甜感物质在TPM基质下甜度值不同程度降低,这可能归因于烟气中其他复杂成分对甜味受体的竞争性或非竞争性抑制作用.②部分物质(葫芦巴内酯和3,5‒二甲基‒1,2‒环戊二酮等)在TPM基质下甜度保持稳定或略有增强,提示烟气基质对不同甜感物质甜度的影响存在差异,甚至可能存在协同效应.这表明在卷烟风味调控中,应重点关注在烟气基质下甜度表现稳定或具有增强效应的成分.

To overcome the limitations of current sweetness measurement methods for cigarette smoke and achieve objective quantification of sweet sensory sensation,HEK293 cell lines with the overexpression of human sweet taste receptor T1R2/T1R3 were established.By employing calcium ion imaging technology to monitor real-time changes in intracellular calcium signals induced by sweet substances,quantitative characterization of sweetness response was realized.Using this technique,sweetness values of ten previously reported representative sweet substances in cigarette smoke including furfural and furanone were systematically determined.Furthermore,the influence of the total particulate matter(TPM)on the sweetness performance of these sweet substances was investigated.The results showed that:1)Most sweet substances exhibited varying degrees of reduced sweetness values in TPM matrix,which may be attributed to the competitive or non-competitive inhibition on the sweet receptors by other complex smoke components.2)Partial substances(fenugreek lactone and 3,5-dimethyl-1,2-cyclopentanedione)maintained stable or slightly enhanced sweetness in the TPM matrix,suggesting that the smoke matrix exerted differential effects on sweetness of different sweet substances and might even involve synergistic effects.These findings suggest that cigarette flavor regulation should focus on components that exhibit stable or enhanced sweetness in the smoke matrix.
