目的 观察植物乳植杆菌 LP45 代谢产物对间充质干细胞增殖、迁移及分化的影响,并基于氧化应激-含 Src 同源2 结构域蛋白酪氨酸磷酸酶(SHP2)/磷脂酰肌醇3-激酶(PI3K)信号通路探讨其作用机制.方法 取对数生长期大鼠骨髓间充质干细胞进行实验,空白组常规培养,LPS 组加入 LPS 培养,LP45 组加入 LPS 和 LP45 上清液培养.CCK-8 法检测细胞增殖能力,Tran-swell 实验检测细胞迁移能力,ELISA 法检测细胞中超氧化物歧化酶(SOD)和丙二醛(MDA)水平,Western blot 法检测细胞中烟酰胺腺嘌呤二核苷酸磷酸氧化酶(NOX)2、NOX4、p-SHP2、p-PI3K、SRY 相关高迁移率蛋白转录因子9(SOX9)、Ⅱ型胶原(ColⅡ)蛋白表达情况.结果 与空白组比较,LPS 组细胞增殖吸光度值明显降低(P<0.05),迁移细胞数量明显减少(P<0.05);细胞中MDA 水平和 NOX2、NOX4 蛋白相对表达量均明显升高(P 均<0.05),SOD 水平和 p-SHP2、p-PI3K、SOX9、ColⅡ蛋白相对表达量均明显降低(P 均<0.05).与 LPS 组比较,LP45 组细胞增殖吸光度值明显升高(P<0.05),迁移细胞数量明显增多(P<0.05);细胞中 MDA 水平和 NOX2、NOX4蛋白相对表达量均明显降低(P 均<0.05),SOD 水平和 p-SHP2、p-PI3K、SOX9、ColⅡ蛋白相对表达量均明显升高(P 均<0.05).结论 植物乳植杆菌 LP45 代谢产物可抑制氧化应激对 SHP2/PI3K 信号通路的抑制作用,促进间充质干细胞的增殖、迁移和分化.

Objective It is to the effect of Lactobacillus plantarum LP45 on the proliferation,migration and differentia-tion of mesenchymal stem cells,and to explore its mechanism of action based on oxidative stress-src homology 2 domain-containing protein tyrosine phosphatase 2(SHP2)/phosphoinositide 3-kinase(PI3K)pathway.Methods Rat bone marrow mesenchymal stem cells in logarithmic growth phase were taken for the experiment.The control group was normally cul-tured,the LPS group was cultured with LPS,and the LP45 group was cultured with LPS and LP45 supernatant.The cell proliferation was detected by CCK-8 assay,the cell migration was detected by Transwell assay,the levels of superoxide dis-mutase(SOD)and malondialdehyde(MDA)in the cells were measured by ELISA,and the protein expressions of nicotin-amide adenine dinucleotide phosphate oxidase(NOX)2,NOX4,p-SHP2,p-PI3K,SRY-related high-mobility protein transcription factor 9(SOX9),and type Ⅱ collagen(Col Ⅱ)in the cells were detected by Western blot.Results Com-pared with the control group,the cell proliferation absorbance value and the number of migrating cells were significantly re-duced in the LPS group(both P<0.05);the level of MDA and the relative protein expressions of NOX2 and NOX4 in the cells were all significantly increased(all P<0.05),while the level of SOD and the relative protein expressions of p-SHP2,p-PI3K,SOX9 and Col Ⅱ in the cells were all significantly reduced(all P<0.05).Compared with the LPS group,the cell proliferation absorbance value and the number of migrating cells were significantly increased in the LP45 group(both P<0.05);the level of MDA and the relative protein expressions of NOX2 and NOX4 in the cells were all significantly de-creased(all P<0.05),while the level of SOD and the relative protein expressions of p-SHP2,p-PI3K,SOX9 and Col Ⅱin the cells were all significantly increased(all P<0.05).Conclusion Lactobacillus plantarum LP45 metabolites can in-hibit the inhibiting effect of oxidative stress on the SHP2/PI3K signaling pathway and promote the proliferation,migration and differentiation of mesenchymal stem cells.
