'云烟87'上部叶成熟进程中萜类香气前体物质的广泛靶向代谢组学分析OA
Widely Targeted Metabolomic Analysis of Terpenoid Aroma Precursors during the Maturation of Upper Leaves in'Yunyan 87'Tobacco
为解析清香型烤烟上部叶成熟进程中萜类香气前体物质的代谢规律,以'云烟 87'上部叶为材料,采用 UPLC-MS/MS 广泛靶向代谢组学技术,系统分析了其成熟早期(T1)、成熟中期(T2)、成熟晚期(T3)的代谢物动态变化规律,重点针对萜类物质进行差异代谢及KEGG 富集分析,共鉴定出 2 441 种代谢物,其中萜类占比最高(495种,占 20.28%).烤烟上部叶成熟进程中萜类物质总体呈"先降后升"趋势:T1 至 T2 阶段萜类物质下调数量(90 个)多于上调数量(39 个),差异组分显著富集于类胡萝卜素生物合成通路,而T2 至T3 阶段萜类物质上调数量(46 个)多于下调数量(20 个),差异组分富集于植物激素信号转导通路,特别是脱落酸(ABA)介导的衰老进程.综上,本研究阐明了'云烟 87'上部叶成熟进程中物质代谢变化规律,揭示 T2 阶段是上部叶萜类香气前体物质形成与转化的关键代谢转折点,为上部叶的适时采收提供了重要的代谢组学依据.
To elucidate the metabolic patterns of terpenoid aroma precursors during the maturation process of upper leaves in fresh-flavor style flue-cured tobacco,upper leaves of'Yunyan 87'were used as the experimental material,and widely targeted metabolomics based on UPLC-MS/MS was applied to systematically analyze the dynamic changes of metabolites at the early(T1),middle(T2),and late(T3)stages of maturation.The analysis specifically focused on the differential accumulation and KEGG pathway enrichment of ter-penoid compounds.A total of 2 441 metabolites were identified,among which terpenoids accounted for the highest proportion(495 compounds,20.28%).During the maturation of upper leaves,terpenoids generally showed a trend of decreasing first and then increas-ing.From T1 to T2,the number of down-regulated terpenoids(90 metabolites)was higher than that of up-regulated ones(39 metabolites),and the differentially accumulated metabolites were significantly enriched in the carotenoid biosynthesis pathway.Con-versely,from T2 to T3,up-regulated terpenoids(46 metabolites)exceeded down-regulated ones(20 metabolites),with the DAMs pri-marily enriched in plant hormone signal transduction pathways,particularly the abscisic acid(ABA)-mediated senescence process.In conclusion,this study elucidates the metabolic shifts during the maturation of upper leaves in'Yunyan 87'and reveals that the T2 stage is a key metabolic turning point for the formation and transformation of terpenoid aroma precursors,providing important metabolomic evi-dence for the timely harvesting of upper leaves.
