消痰散结方对结直肠癌细胞上皮间质转化和化疗耐药性的影响机制研究OA
Effect of Xiaotan Sanjie Formula(消痰散结方)on epithelial-mesenchymal transition and che-moresistance in colorectal cancer cells
目的 探讨消痰散结方调节黏着斑激酶(FAK)/丝裂原活化细胞外信号调节激酶(MEK)/细胞外信号调节激酶(ERK)通路对结直肠癌细胞上皮-间质转化(EMT)和化疗耐药性的影响.方法 MTT法筛选消痰散结方最佳浓度.HCT116细胞分为对照组,L、M、H-消痰散结方组(1.24、2.48、3.74 mg/L)、消痰散结方+ML-099组(20 μmol/L MEK/ERK通路激活剂ML-099+3.74 mg/L消痰散结方);HCT116/5-氟尿嘧啶细胞分为HCT116/5-氟尿嘧啶组、HCT116/5-氟尿嘧啶+消痰散结方组(3.74 mg/L).检测细胞增殖、迁移、侵袭、EMT及通路相关蛋白.结果 L、M、H-消痰散结方组较对照组HCT116细胞划痕愈合率、侵袭细胞数、FAK、MEK、ERK、Vimentin、N-cadherin降低,E-cadherin升高(P<0.05);消痰散结方+ML-099组较H-消痰散结方组上述改变被部分逆转.与耐药组比较,消痰散结方处理后HCT116/5-氟尿嘧啶细胞IC50值、增殖活性、划痕愈合率、侵袭细胞数及FAK、MEK、ERK、Vimentin、N-cadherin降低,E-cadherin升高(P<0.05).结论 消痰散结方通过抑制FAK/MEK/ERK通路抑制结直肠癌细胞增殖、迁移、侵袭及EMT,提高结直肠癌细胞对5-氟尿嘧啶的化疗敏感性.
Objective To investigate the effect of Xiaotan Sanjie Formula(消痰散结方,XTSJF)on epithelial-mesenchymal transition(EMT)and chemoresistance in colorectal cancer cells by regulating the focal adhesion kinase(FAK)/mitogen-activated extracellular signal-regulated kinase(MEK)/extracellular signal-regulated kinase(ERK)signaling pathway.Methods MTT assay was used to screen the optimal concentration of XTSJF.HCT116 cells were grouped into control group,L,M,H-XTSJF group(1.24、2.48、3.74mg/L)and XTSJF+ML-099 group(20 μmol/L MEK/ERK pathway activator ML-099+3.74 mg/L XTSJF);HCT116/5-fluorouracil cells were separated into HCT116/5-fluorouracil group and HCT116/5-fluorouracil+XTSJF group(3.74mg/L).The proliferation,migration,inva-sion,EMT,and pathway related proteins of HCT116 and HCT116/5-fluorouracil cells were detected.Results Compared with the control group,the scratch healing rate,number of invasive cells,FAK,MEK,ERK,Vimentin,and N-cadherin protein expression of HCT116 cells in the L,M,and H-XTSJF group were all reduced,the expression of E-cadherin protein increased(P<0.05),Compared with the H-XTSJF group,the above changes were partially reversed in the XTSJF+ML-099 group.Compared with the drug-resistant group,XTSJF treatment significantly reduced the IC50 value,proliferative activity,wound healing rate,number of invasive cells,and protein expression levels of FAK,MEK,ERK,Vimentin,and N-cadherin in HCT116/5-FU cells,while E-cadherin expression was signifi-cantly increased(P<0.05)..Conclusion XTSJF inhibits the proliferation,migration,invasion,and EMT of colorectal cancer cells by inhibiting the FAK/MEK/ERK pathway,thereby increasing the chemotherapy sensitivity of colorectal cancer cells to 5-fluorouracil.
