目的 探讨雷公藤多苷(GTW)调控AHR信号通路对膜性肾病(MN)大鼠肾组织氧化应激和炎症的影响及其潜在机制.方法 将清洁级雄性SD大鼠,随机选取8只为空白组,其余27只大鼠均采用单次尾静脉注射羊抗大鼠Fx1A血清造模构建MN模型,MN模型大鼠复制成功后随机分为模型组、盐酸贝那普利组、雷公藤多苷组,连续灌胃6周.收集大鼠24h尿液及腹主动脉血液进行生化检测;利用HE染色和PASM染色观察大鼠肾组织病理形态;采用ELISA检测大鼠肾脏血清中超氧化物歧化酶(SOD)、丙二醛(MDA)表达情况;RT-PCR法和Western blot法检测大鼠肾组织中炎症及氧化应激通路相关因子AHR、NF-κB p65、COX-2、Nrf2 mRNA及蛋白表达水平.结果 GTW明显降低MN大鼠24h-UTP(P<0.01)、血清BUN、Cr、TC、TG水平(P<0.01),降低肾组织MDA含量(P<0.01),提高血清ALB水平及肾组织中SOD活性含量(P<0.01),改善MN大鼠肾组织损伤,降低肾组织AHR、NF-κB p65、COX-2 mRNA及蛋白表达水平(P<0.01),提高Nrf2 mRNA及蛋白表达水平(P<0.01).结论 GTW可能通过抑制AHR信号通路,减轻炎症反应、缓解肾组织氧化应激,从而降低尿蛋白,改善肾功能、延缓疾病进展.

Objective To investigate the mechanism of glycosides of Tripterygium wilfordii(GTW)in alleviating oxidative stress and inflammation in renal tissue of rats with membranous nephropathy(MN)by modulating the aryl hydrocarbon receptor(AHR)signaling pathway.Methods Thirty-five SPF male SD rats were used.Eight served as the blank control group.The remaining 27 were used to establish the MN model via a single tail vein injection of sheep anti-rat Fx1A serum.After successful model confirmation,these rats were randomly divided into three groups(n=9 each):the MN model group,the benazepril hydrochloride group[10 mg/(kg·/d)],and the GTW group(6.25 mg/kg/d).All groups received daily intragastric administration for 6 weeks.We collected 24-hour urine to mea-sure total urinary protein(24h-UTP).Blood samples from the abdominal aorta were analyzed for serum albumin(ALB),aspartate ami-notransferase(AST),alanine aminotransferase(ALT),urea nitrogen(BUN),creatinine(Cr),total cholesterol(TC),and triglyceride(TG).Renal histopathology was assessed by hematoxylin-eosin(HE)and periodic acid-Schiff methenamine(PASM)staining.The levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in renal tissues were measured by enzyme-linked immunosorbent assay(ELISA).The mRNA and protein expression levels of AHR,NF-κB p65,COX-2,and Nrf2 in renal tissues were determined by RT-PCR and Western blot(WB),respectively.Results Compared with the model group,GTW treatment significantly decreased 24h-UTP,serum BUN,Cr,TC,TG,and renal MDA content(all P<0.01),while it increased serum ALB and renal SOD activity(P<0.01).GTW also ameliorated renal histopathological damage.Furthermore,GTW downregulated the mRNA and protein expression of AHR,NF-κB p65,and COX-2,and upregulated Nrf2 expression in renal tissues(all P<0.01).Conclusion GTW likely exerts its renoprotective effects in MN by inhibiting the AHR signaling pathway,which subsequently attenuates inflammatory response and oxida-tive stress,leading to reduced proteinuria,improved renal function,and delayed disease progression.
