补阳还五汤调控细胞自噬改善HPH肺血管重构的机制研究OA
Mechanism Study on Buyang Huanwu Decoction(补阳还五汤)in regulating autophagy to improve pulmonary vascular remodeling in hypoxic pulmonary hypertension
目的 探讨补阳还五汤(BHD)调控细胞自噬改善低氧性肺动脉高压(HPH)肺血管重构的作用机制.方法 将60只SPF级雄性SD大鼠随机分为空白对照组、模型组、西地那非组及补阳还五汤低、中、高剂量组,每组10只.除空白对照组外,其余各组采用常压间断缺氧法构建HPH模型,同时给予相应药物干预.检测RVSP、RVHI,HE染色观察肺血管结构,Western Blot和PT-PCR检测Beclin-1、ATG5、P62、PCNA以及PI3K/AKT/mTOR信号通路蛋白表达.结果 与空白对照组比较,HPH模型组RVSP、RVHI显著升高,肺小动脉壁增厚、肌化率增加,PCNA、PI3K、AKT、mTOR表达上调,Beclin-1、ATG5表达上调、P62表达下降(P<0.01).与模型组比较,补阳还五汤各组及西地那非组上述指标均显著改善,且补阳还五汤的作用呈剂量依赖性(P<0.01).结论 补阳还五汤能通过抑制PI3K/AKT/mTOR信号通路激活细胞自噬,减少肺动脉平滑肌细胞过度增殖,从而改善HPH肺血管重构.
Objective To investigate the mechanism by which Buyang Huanwu Decoction(补阳还五汤,BHD)improves pulmonary vas-cular remodeling in hypoxic pulmonary hypertension(HPH)through regulating autophagy.Methods Sixty SPF male SD rats were ran-domly divided into a blank control group,a model group,a sildenafil group,and low-,medium-,and high-dose BHD groups,with 10 rats in each group.Except for the blank control group,the HPH model was established in the other groups using the normobaric intermit-tent hypoxia method,and corresponding drug interventions were administered simultaneously.The right ventricular systolic pressure(RVSP)and right ventricular hypertrophy index(RVHI)were detected;pulmonary vascular structure was observed by HE staining;the protein expressions of Beclin-1,ATG5,P62,PCNA and PI3K/AKT/mTOR signaling pathway-related proteins were detected by West-ern Blot and RT-PCR.Results Compared with the blank control group,the RVSP and RVHI in the HPH model group were significantly increased,the pulmonary arteriole wall was thickened with an elevated muscularization rate,the expressions of PCNA,PI3K,AKT and mTOR were up-regulated,the expressions of Beclin-1 and ATG5 were up-regulated,and the expression of P62 was down-regulated(P<0.01).Compared with the model group,all the above indicators were significantly improved in each BHD group and the sildenafil group,and the effect of BHD was dose-dependent(P<0.01).Conclusion BHD can activate autophagy by inhibiting the PI3K/AKT/mTOR signaling pathway,reduce the excessive proliferation of pulmonary artery smooth muscle cells,and thereby improve pulmonary vascular remodeling in HPH.
