CCO基因家族成员广泛存在于细菌、植物和动物中,在植物次生代谢中起关键作用.本研究通过生物信息学方法鉴定得到了18个辣椒CCO基因家族成员,命名为CaCCO01-CaCCO18,分布于第1、3、5、6、7、8、10、11号共8条染色体上,氨基酸序列长度范围为102~646 aa,分子质量范围为11 224.87~72 718.77 Da,等电点介于 5.13~9.19.系统发育分析将其划分为 6 个亚群,9 个基因属于 CCD 亚家族,4 个基因属于NCED 亚家族,另有 5 个基因(CaCCO11、CaCCO15、CaCCO16、CaCCO17、CaCCO18)可能划分为CCDL亚家族.顺式作用元件分析发现,所有CaCCO基因均检测到光响应、激素响应、生长发育及胁迫响应等顺式作用元件.CaCCO02 含有最多的TATC-box(7个),CaCCO12包含最多的RY-element(18个).转录组测序和表达分析表明,CaCCO02 和CaCCO12呈早期高表达、随发育进程逐渐下调的趋势,CaCCO04表达水平从辣椒开花后12 d(S1)到48 d(S4)持续上升,CaCCO06在开花后36 d(S3)时期特异性高表达,呈现明显的中期表达峰,qRT-PCR 分析与转录组分析结果基本一致,说明这4个基因可能在辣椒果实发育和形态建成中发挥重要的调控作用.

Members of the CCO gene family are ubiquitously distributed in bacteria,plants and animals,and perform pivotal functions in plant secondary metabolism.In this study,a total of 18 CCO family genes were identified in Capsicum annuum via bioinformatic approaches,designated CaCCO01-CaCCO18.These genes were mapped to eight chromosomes(Chr.1,3,5,6,7,8,10 and 11).The encoded amino acid sequences ranged from 102 to 646 amino acids(aa),with molecular weights varying from 11224.87 to 72718.77 Da and isoelectric points(pI)ranging between 5.13 and 9.19.Phylogenetic clustering classified these CaCCOs into six subgroups:nine genes belonged to the CCD subfamily,four to the NCED subfamily,and the remaining five genes(CaCCO11,CaCCO15,CaCCO16,CaCCO17,CaCCO18)were tentatively assigned to the CCDL subfamily.Cis-acting element prediction revealed cis-regulatory motifs associated with light responses,phytohormone signaling,growth and development,as well as stress responses across all CaCCO promoters.CaCCO02 harboured the largest number of TATC-box elements(7 copies),while CaCCO12 contained the highest abundance of RY-element motifs(18 copies).Transcriptome sequencing and expression profiling demonstrated distinct spatiotemporal expression patterns for key CaCCO genes:CaCCO02 and CaCCO12 exhibited high expression at early fruit developmental stages followed by gradual downregulation as fruit maturation proceeded;transcript abundance of CaCCO04 increased continuously from 12 days post-anthesis(DPA,stage S1)to 48 DPA(stage S4);CaCCO06 displayed stage-specific peak expression at 36 DPA(stage S3),representing a prominent mid-developmental expression maximum.Quantitative real-time PCR(qRT-PCR)validation yielded results largely consistent with transcriptome data,indicating that these four genes likely act as critical regulators governing fruit development and morphogenesis in pepper.
