毛竹叶总黄酮的超声波辅助酶法提取及其成分分析OA
Ultrasound-assisted Enzymatic Extraction and Component Analysis of Total Flavonoids From Moso Bamboo Leaves
以毛竹竹叶为原料,采用超声波辅助酶法提取毛竹叶总黄酮,以体积分数60%乙醇为提取溶剂,在单因素试验的基础上利用响应面法优化工艺条件.研究结果表明:1 g 的毛竹竹叶粉末最优提取条件为纤维素酶添加量1.4%(以竹叶质量计)、乙醇和原料液固比34∶1(mL∶g)、提取温度55℃、提取时间40 min、超声波功率336 W,该条件下竹叶总黄酮得率为2.460%.采用扫描电镜(SEM)、傅里叶红外光谱(FT-IR)及纳米粒度仪对提取前后的毛竹竹叶进行了表征,分析结果显示:经超声波辅助酶法处理后,酶解后的竹叶粉末表面更粗糙,竹叶细胞壁结构遭到破坏,促进了竹叶黄酮类物质从细胞内向提取液的转移,从而提高了竹叶总黄酮的提取得率.通过对比乙醇处理、超声波处理、酶解处理和超声波辅助酶解处理这4 种方法,发现采用超声波辅助酶法提取的竹叶总黄酮得率为 2.465%、总多酚得率为10.874%以及 DPPH 自由基清除率为87.791%,均最高.高效液相色谱(HPLC)分析表明:超声波辅助酶法处理的竹叶提取液中多种黄酮类化合物(牡荆苷、异牡荆苷、荭草苷、异荭草苷及芹菜素),含量均高于其他处理组.
Moso bamboo(Phyllostachys edulis(Carrière)J.Houz)leaves were used as raw material to extract flavonoids via ultrasound-assisted enzymatic method.Using 60%ethanol as the extraction solvent,the process conditions were optimized by response surface method on the basis of single-factor test.The results showed that the optimal extraction conditions for 1 g P.edulis leaves powder were as follows:cellulose addition of 1.4%(based on the mass of bamboo leaves),liquid-solid ratio of 34∶1(mL∶g),extraction temperature of 55℃,extraction time of 40 min,and ultrasound power of 336 W.Under these conditions,the total flavonoid yield from bamboo leaves reached 2.460%.The P.edulis leaves before and after extraction were characterized by scanning electron microscopy(SEM),Fourier transform infrared spectroscopy(FT-IR)and nanoparticle size analyzer.Analysis results revealed that ultrasound-assisted enzymatic treatment resulted in a rougher surface of the enzymatically treated material and disrupted cell wall structures,facilitating the transfer of flavonoids from the cells into the extraction solution and thereby enhancing extraction efficiency.By comparing four methods of ethanol treatment,ultrasound treatment,enzymatic hydrolysis,and ultrasound enzymatic hydrolysis,it was found that the yield of total flavonoid,the yield of total polyphenol and antioxidant activity of bamboo leaves flavonoid extracted by ultrasound-assisted enzymatic hydrolysis were 2.465%,10.874%and 87.791%,respectively,which were the highest.High performance liquid chromatography(HPLC)analysis revealed that the contents of various flavonoids(vitexin,isovitexin,orientin,isoorientin and apigenin)in bamboo leaves extract treated by ultrasound-assisted enzymatic method were significantly higher than those in other treatment groups.
