贵州4个地方黄牛品种(关岭牛、思南牛、威宁牛和务川黑牛)是西南地区重要的遗传资源,但其母系遗传背景尚缺乏基于线粒体全基因组数据的系统评估.本研究对上述4个品种共80个个体的线粒体DNA全基因组进行测序、组装,并与11条已知家牛单倍型参考序列进行比对,构建系统发育树和单倍型网络图.结果表明,在80个个体中共检测到68种mtDNA全基因组单倍型、441个变异位点;单倍型多样度为0.990±0.006,核苷酸多样度为0.007 9±0.001 4,平均核苷酸差异数为128.4,表明贵州4个地方黄牛品种均具有丰富的母系遗传多样性.系统发育分析显示,普通牛(T支系)占52.50%(42/80),瘤牛(I支系)占46.25%(37/80),稀有原牛Q支系占1.25%(1/80);其中普通牛以T3支系为主,瘤牛以I1a亚支系为主.上述结果揭示,贵州4种地方黄牛均为普通牛与瘤牛的混合母系起源,群体结构相对均一;务川黑牛中发现的稀有Q支系为中国地方黄牛中罕见的原始血统残留,具有重要的保种价值.

Four local cattle breeds in Guizhou Province(Guanling cattle,Sinan cattle,Weining cattle,and Wuchuan black cattle)are important genetic resources in the Southwest region of China.However,their maternal genetic background has not yet been systematically evaluated using complete mitochondrial ge-nome data.In this study,complete mitochondrial DNA whole genomes were sequenced and assembled for 80 individuals from these four breeds.The sequences were compared with 11 known cattle haplotype refe-rence sequences,and phylogenetic trees and haplotype networks were constructed.A total of 68 complete mtDNA haplotypes and 441 variable sites were identified among the 80 individuals.The haplotype diversity was 0.990±0.006,the nucleotide diversity was 0.007 9±0.001 4,and the average number of nucleotide differences was 128.4,indicating abundant maternal genetic diversity in the four local cattle breeds of Guizhou.Phylogenetic analysis showed that the taurine lineage(T lineage)accounted for 52.50%(42/80),the indicine lineage(I lineage)accounted for 46.25%(37/80),and the rare aurochs Q lineage accounted for 1.25%(1/80).The taurine ancestry was mainly represented by the T3 lineage,whereas the indicine ances-try was dominated by the I1a sublineage.These results indicate that the four local cattle breeds in Guizhou have mixed maternal origins from both taurine and indicine cattle,with a relatively uniform population structure.The rare Q lineage detected in Wuchuan black cattle represents a rare remnant of ancient mater-nal ancestry in Chinese local cattle and has important conservation value.
