牛坏死杆菌43K OMP对鼠乳腺上皮细胞分泌炎性因子的影响OA
Effects of 43KDa OMP from Fusobacterium necrophorum on Inflammatory Cytokines Secretion in Mouse Mammary Epithelial Cells
为了研究牛坏死杆菌外膜蛋白 43 kDa(Out membrane protein of 43 kDa,43K OMP)对鼠乳腺上皮细胞(MMECs)分泌炎性因子的影响,利用重组 43K OMP 刺激 MMECs,通过检测与炎症相关的细胞因子来探究牛坏死杆菌的免疫作用机制.利用CCK-8 筛选在 10、20 和 40 μg·mL-1 不同浓度下 43K OMP 作用在 MMECs 不同时间点的细胞抑制率.以筛选最佳浓度的43K OMP 刺激MMECs,分别于 6,12,18 和24 h 不同时间收集样品,利用 qRT-PCR 检测 MMECs 炎性细胞因子 IL-6、IL-1β 和TNF-α 的变化,应用 ELISA 方法检测细胞上清中的 IL-6、IL-1β 和 TNF-α 等炎性细胞因子的分泌水平.结果表明:43K OMP刺激MMECs 细胞浓度为20 μg·mL-1,作用时间为 24 h.20 μg·mL-1 浓度下 43K OMP 刺激 MMECs 18 h 和 24 h 时,细胞的 IL-6 mRNA、IL-1β mRNA 和 TNF-α mRNA 相对表达水平极显著升高(P<0.01);43K OMP 刺激 MMECs 24 h,细胞上清中 IgG、IL-6、IL-1β、IL-8 及 TNF-α 分泌量显著升高(P<0.05),其他时间点,细胞上清中 IgG、IL-6、IL-1β、IL-8 及 TNF-α 分泌量呈现不同程度的升高趋势.结果表明:牛坏死杆菌外膜蛋白 43K OMP 能促进 MMECs IL-6、IL-1β和 TNF-α的分泌,促进细胞分泌多种促炎性细胞因子,在牛坏死杆菌病引起的免疫防御反应中起到一定的作用.
To investigate the effects of 43 kDa outer membrane protein(43K OMP)derived from Fusobacterium necrophorum on the secretion of inflammatory factors in mouse mammary epithelial cells(MMECs),and to clarify the immune mechanism of Fusob-acterium necrophorum by detecting inflammation-related cytokines after recombinant 43K OMP stimulation of MMECs.The CCK-8 assay was used to screen the cell inhibition rates of MMECs treated with 43K OMP at concentrations of 10,20 and 40 μg·mL-1 for different durations.MMECs were stimulated with the optimal concentration of 43K OMP,and samples were collected at 6,12,18,and 24 h post-treatment.The mRNA relative expression of inflammatory cytokines IL-6,IL-1β and TNF-α was determined via qRT-PCR,while the secretion levels of these cytokines in cell supernatants were quantified by ELISA.These results showed that the optimal stimulation condition was 20 μg·mL-1 43K OMP with a treatment duration of 24 h.When MMECs were incubated with 20 μg·mL-1 43K OMP for 18 h and 24 h,the relative mRNA expression of IL-6,IL-1β and TNF-α increased extremely significantly(P<0.01).After 24 h of stimulation,the contents of IgG,IL-6,IL-1β,IL-8 and TNF-α in cell supernatants were significantly elevated(P<0.05),and varying degrees of increasing trends were observed at other time points.In conclusion,43K OMP of F.necrophorum can accelerate the secretion of IL-6,IL-1β and TNF-α in MMECs and induce the production of multiple pro-inflammatory cytokines,which participates in host immune defense against F.necrophorum infection.
