Effects of ADSCs on Mitochondrial Biogenesis in Canines with Acute Pancreatitis

为探究脂肪干细胞(Adipose-derived stem cells,ADSCs)对犬急性胰腺炎(Acute pancreatitis,AP)的治疗效果,将 9 只中华田园犬随机分为 CON 组、AP 组和 ADSCs 组.于十二指肠大乳头肠系膜对侧切开肠壁,向 AP 组和 ADSCs 组犬的胰胆管中注射牛磺胆酸钠(5%,0.1 mL·kg-1)+胰蛋白酶(3 500 U·kg-1),CON 组推注等量生理盐水.ADSCs 组术后静脉注射 ADSCs(1×106 个·kg-1)进行治疗,CON 组和 AP 组注射生理盐水.术后 24 h 采集血液和胰腺组织样本,检测临床体征、疼痛评分、胃蛋白酶、胰蛋白酶、ATP、Ca-ATP 酶及线粒体生物合成标志物的表达水平.结果显示,各组犬术中及术后生理指标稳定;AP 组胃蛋白酶和胰蛋白酶活性显著升高,Ca-ATP 酶活性、ATP 含量及线粒体生物合成标志物表达水平显著降低;ADSCs 治疗可显著降低胃蛋白酶和胰蛋白酶活性,并上调线粒体生物合成标志物表达水平.结果表明,采用手术法建立的犬急性胰腺炎模型安全性良好,脂肪干细胞可通过抑制胃蛋白酶和胰蛋白酶分泌、增强线粒体生物合成能力,从而发挥胰腺保护作用.研究可为急性胰腺炎的发病机制研究和脂肪干细胞的临床应用提供实验基础.

To investigate the therapeutic efficacy of adipose-derived stem cells(ADSCs)against acute pancreatitis(AP)in canines,nine native Chinese rural canines were randomly divided into the control(CON)group,AP group,and ADSCs group.An incision was performed on the intestinal wall opposite the mesentery of the major duodenal papilla.The AP and ADSCs groups were subjected to retrograde pancreaticobiliary infusion of 5%sodium taurocholate(0.1 mL·kg-1)and trypsin(3 500 U·kg-1),while the CON group received an identical volume of sterile normal saline.Postoperatively,canines in the ADSCs group were intravenously administrated ADSCs at 1×106 cells·kg-1 for intervention,and the CON and AP groups were injected with equal-volume normal saline.Blood and pancreatic tissue samples were collected at 24 h post-operation to detect clinical manifestations,pain scores,pepsin and trypsin activities,ATP content,Ca-ATPase activity and the expression of mitochondrial biogenesis markers.The results showed that physiological indicators of all experimental canines remained stable intraoperatively and postoperatively.In the AP group,the activities of pepsin and typsin were significantly increased,while Ca-ATPase,ATP concentration and the expression levels of mitochondrial biogenesis markers were markedly decreased.ADSCs treatment significantly reduced elevated pepsin and trypsin activities and upregulated the expression of mitochondrial biogenesis markers.In conclusion,the surgically established canine acute pancreatitis model has reliable safety.ADSCs protect pancreatic tissues via restraining pepsin and trypsin secretion and enhancing mitochondrial biogenesis,which provides experimental data for exploring AP pathogenesis and clinical application of ADSCs.
